摘要
目的探讨miR-17-92基因簇在食管鳞癌中的表达,并分析其与肿瘤分化程度及预后的关系。方法半定量RT-PCR和实时荧光定量PCR方法分别检测58例ESCC组织和来自同一患者的邻近正常食管组织中miR-17-92的表达。采用Mann Whitney-U非参数检验和卡方检验比较低表达组和高表达组的临床病理特征和预后指标,采用Kaplan-Meier法分析miR-17-92表达水平与食管鳞癌患者生存时间之间的关系。结果 miR-17-92在所有的组织中都有表达,且在食管鳞癌组织中miR-17-92的表达高于正常食管组织(P<0.001)。miR-17-92在TNM分期越高、分化程度越低的肿瘤组织中表达越高(分别是P=0.032,P=0.035)。另外,miR-17-92高表达的患者发生转移和死亡的风险明显增高,预后更差(P=0.038)。结论 miR-17-92基因簇在食管鳞癌组织和邻近正常食管组织中表达存在差异性,可能在食管鳞癌发生、发展过程中起重要作用,有望成为食管鳞癌患者预后一种新的指标。
Objective To investigate the expression of microRNA-17-92(miR-17-92)in esophageal squamous carcinoma(ESCC)and the relationship between miR-17-92 and clinicopathological features.Methods Expression levels of miR-17-92in58 pairs of ESCC and adjacent normal tissues were detected by semi-quantitative RT-PCR and real-time quantitative RT-PCR.Mann Whitney-U nonparametric test and chi-square test were performed to determine the clinicopathological features and prognosis of higher miR-17-92 expression group and lower miR-17-92 expression group.Then,Kaplan-Meier analyses were performed to determine the association of miR-17-92 expression with survival of ESCC patients.Results miR-17-92 expression in ESCC tissues was significantly higher than that in adjacent normal tissues(P〈0.001).There was a correlation between miR-17-92 upregulation and advanced TNM stage(P=0.032).There was a correlation between miR-17-92 upregulation and lower differentiated histologic grade of ESCC patients(P=0.035).In addition,ESCC patients with higher miR-17-92 expression had significantly poorer overall survival(P=0.038).Conclusion The expression of miR-17-92 cluster closely correlates with the occurrence and progress of esophageal squamous carcinoma and may be used as an indicator for esophageal squamous carcinoma prognosis.
