摘要
目的通过观察三七总皂甙对脑缺血-再灌注大鼠海马区血管内皮生长因子(VEGF)表达的影响,探讨三七总皂甙(PNS)对脑缺血-再灌注大鼠发挥脑保护作用可能的相关机制。方法采用改良的线栓法建立大脑中动脉闭塞脑缺血-再灌注大鼠模型,随机将60只健康雄性Wistar大鼠分为:假手术组、模型组和PNS治疗组。应用神经行为学评分评价大鼠神经功能症状,免疫组织化学法检测大鼠海马区VEGF的阳性表达情况。结果模型组与PNS治疗组比较脑缺血-再灌注大鼠的神经功能评分显著提高(P<0.01);PNS治疗组VEGF阳性表达较假手术组和模型组均明显增高(均P<0.01)。结论 PNS的治疗干预可以使缺血再灌注模型大鼠的行为学症状得到明显改善,其机制可能是通过促进脑缺血-再灌注大鼠脑组织VEGF表达,抑制神经细胞凋亡,发挥神经保护作用。
Objective To observe the effect of the Panax Notoginseng saponins on the expression of vascular endothelial growth factor( VEGF) in hippocampus of rats with cerebral ischemia reperfusion injury,and to explore the possible mechanism of the protective effect of the Panax Notoginseng saponins( PNS) on cerebral ischemia reperfusion injury rats. Methods A rat model of cerebral ischemia and reperfusion was established by using the modified thread embolism method. 60 healthy male Wistar rats were randomly divided into three groups:sham operation group,model group and PNS treatment group. The positive expression of VEGF in the hippocampus of rats was detected by using the neurological behavior score. Results The neurological function score of model group compared to the PNS treatment group was significantly improved( P〈0. 01); the expression of VEGF in PNS group was significantly higher than that in sham operation group and model group( P〈0. 01). Conclusion The therapeutic intervention of PNS can improve the behavior of the model rats. The mechanism may be to inhibit the apoptosis of nerve cells and to play a neuroprotective protective effect by means of improving the expression of VEGF in brain tissue of rats with cerebral ischemia reperfusion injury.
