迷走神经电刺激保护失血性休克肺微血管内皮细胞 被引量:2

Vagal Stimulation Attenuates Hemorrhagic Shock-Induced Pulmonary Microvascular Endothelial Cell Injury

摘要 目的:观察电刺激迷走神经对失血性休克大鼠肺微血管内皮细胞的影响。方法:成年雄性SD大鼠30只,体质量240-260g,随机分为3组:假休克组(SS组)、失血性休克组(HS组)、迷走神经电刺激组(STM组),每组10只。Wiggers改良法制备失血性休克模型,模型制备成功3h后留取血和肺组织标本。光镜下观察肺组织形态学变化,电镜下观察肺微血管内皮细胞形态改变,Western印记法测Src抑制的蛋白激酶C的底物(SSeCKS)蛋白表达、ELISA法测血浆肿瘤坏死因子-α(TNF-α)浓度。结果:HS组肺组织和内皮细胞形态较SS组遭到明显破坏,STM组较HS组破坏减轻,接近SS组;HS组SSeCKS表达和TNF-α浓度比SS组明显升高(P<0.05),STM组可减少SSeCKS表达和TNF-α浓度(P<0.05)。结论:电刺激迷走神经对失血性休克大鼠肺微血管内皮细胞可产生保护作用,其机制可能与激活胆碱能抗炎通路拮抗全身炎性反应有关。 Objective:To observe the effects of electrical vagus nerve stimulation on hemorrhagic shockinduced pulmonary microvascular endothelial cell(PMVEC)injury.Methods:Thirty male Sprague-Dawley rats(weighing 240-260g)were randomly divided into three groups:sham shock group(SS group),hemorrhagic shock group(HS group),and vagus nerve stimulation group(STM group),with 10 rats in each group.After the hemorrhagic shock models were built 3h later,all samples of blood and lung tissues were collected.The histological changes of lung tissue and PMVEC were observed respectively using light microscope and transmission electron microscope;Western blotting were used to detect the expression of SSeCKS;the concentration of plasma tumor necrosis factor-α(TNF-α)was detected by using enzyme linked immunosorbent assay.Results:The histological changes of lung tissue and PMVEC were severely damaged in HS group than in SS group;the damages were ameliorated in STM group than in HS group.The expression of SSeCKS and the concentration of TNF-αincreased in HS group versus SS group(P〈0.05)and STM group(P〈0.05).Conclusion:Electrical vagus nerve stimulation could protect PMVEC inhemorrhagic shock rats.Activation of cholinergic anti-inflammatory pathway,and inhibition of inflammatory responses may be involved in the mechanism.
