双孢蘑菇蛋白质提取工艺优化及其电泳分析 被引量:2

Electrophoretic analysis and optimization of protein extraction for mushroom

摘要 用Tris-HCl法提取双孢蘑菇蛋白质,以蛋白质提取率为因变量,以提取液浓度、提取时间、提取液p H、料液比为自变量,通过单因素试验和响应曲面法得到最优工艺参数,并对蛋白质干粉进行裂解及SDS-PAGE电泳,分析双孢蘑菇蛋白质亚基分布及分子量。结果表明,双孢蘑菇蛋白质提取最佳工艺参数为提取液浓度0.2 mol/L、提取时间8 h、提取液p H8.4、料液比1:15,提取率可达75.09%。双孢蘑菇SDS-PAGE分析表明,双孢蘑菇蛋白质中约含19条亚基,主要分布在26.4-80.8 ku。 Tris-HCl was used as the extraction solvent for extracting protein from mushroom.The extraction yield was used as the dependent variable and Tris-HCl concentration,extraction duration,p H,material/liquid ratio was used as the independent variables.The single-factor and response surface methodology was used to optimize the extraction processing of protein from mushroom.The SDS-PAGE was used to analysis the distribution of subunits and ralative molecular mass.The optimal conditions for extracting proteins from mushroom were determined to be that Tris-HCl concentration 0.2 mol/L,extraction duration 8 h,p H8.4 and material/liquid ratio 1:15.A protein yield of 75.09% was obtained under the optimal extraction conditions.The SDS-PAGE analysis showed that the proteins from mushroom contained approximately 19 subunits and that majority of them had a molecular mass varying from 26.4 ku to 80.8 ku.
