摘要
用BALB/C小鼠骨髓瘤细胞(SP2/0)与经基孔肯雅(CHIK)病毒免疫后的BALB/C小鼠的B淋巴细胞进行融合,共获得11株能分泌抗CHIK病毒McAb杂交瘤细胞系。融合率为80.6%,阳性率为29.7%。经微量中和试验证明,11株McAb均具有抗CHIK原型及从棕果蝠与人体分离到的地方株CHIK病毒的中和抗体。血凝抑制试验(HI)及间接免疫荧光试验(IF)均为阳性,McAb培养上清效价前者为1:32~1:64,后者为1:40~1:80,McAb腹水效价前者1:2560~1:5120,后者1:2560~1:10240。11株McAb均为IgG2b亚类,具有明显的病毒特异性。
In this study, we applied lymphocyte hybridoma technic, used the BALB/c mouse myeloma cell lines (SP/O) and the B lymphocytes of BALB/c mouse, finally 11 cell lines of hybridoma which enable to secrets McAb against CHIK virus have been obtained. The fusion rate is 80. 6%, the positive fate is 29. 7%. All the McAb from 11 hybridoma cell lines not only have been identified by neutralization test to have ability against the original form of CHIK virus and also the endemic strains isolated from Rousethus Ieschenautti and patient's blood. The titers of McAb in culture supernatants are 1:32-1:64 in HI test and 1: 40- 1:80 in IF test. The liters of McAb in ascitic fluids are 1:2560- 1:5120 in HI test and 1:2560-1 :10240 in IF lest. All the McAb from 11 cell line of hybridoma have been further comfirmed to sublype IgG2b. All the 11 hybridoma cell line secreled anlibodies slably, possess a higher liter and specificity. When test usually shows a strong specific reaction eilher to freshly isolated virus strains or the original form of CHIK virus.
