拉科酰胺对神经病理性疼痛大鼠的镇痛作用 被引量:1

摘要 目的观察拉科酰胺在大鼠神经病理性疼痛中的镇痛作用。方法制备神经病理性疼痛(DNP)模型大鼠27只,随机分为3组(n=9):A组为腹腔内注射生理盐水(NS)5ml;B组为腹腔内注射拉科酰胺60mg;C组为腹腔内注射拉科酰胺120mg。测定给药前、给药后10、20、30、40、50min大鼠机械性缩足反射阈值(Mwr)和热缩足潜伏期(TwL)。结果各时间点B组大鼠MWT[10min(17.46±0.71)g;20min(18.17±0.61)g;30min(17.21±0.41)g;40min(17.82±0.53)g;50min(18.17±0.37)g],TWL{10min(9.21±0.37)s;20min(8.90±0.62)s:30min(9.46±0.29)s:40min(9.28±0.73)s;50min(9.41±0.32)s],均较A组明显增加(P〈0.05);各时间点C组大鼠MWT[10min(21.02±0.29)g;20min(20.89±0.35)g;30min(20.24±0.62)g;40min(21.71±0.36)g;50min(21.07±0.74)gl,TWL[10min(10.46±0.23)s;20min(11.21±0.74)s;30min(10.97±0.27)S;40min(10.62±0.46)s;50min(10.71±0.39)s],均较A组明显增加(P〈0.01)。结论腹腔内注射拉科酰胺对大鼠神经病理性疼痛有较强的镇痛作用。 Objective To evaluate the analgesic effect of lacosamide on neuropathic pain in rats. Methods Twenty - seven rats were used to establish the models of neuropathic pain. The rats were ran- domly divided into 3 groups ( n = 9 ) : group A, 5 ml normal saline was intraperitoneally injected; group B, 60 mg laeosamide was intraperitoneally given; group C, 120 mg lacosamide was intraperitoneally adminis- tered. The mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) were meas- ured before and 10, 20, 30, 40 and 50 rain after administraton respectirely. Results In group B, MWT at each time point [10min: (17.46±0.71) g; 20 min: (18. 17 ±0.61) g; 30 min: (17.21 ±0.41) g; 40min: (17.82±0.53) g; and 50 min: (18. 17 ±0.37) g], andTWL ±10 min: (9.21 ±0.37) s; 20min: (8.90 ±0.62) s; 30 min: (9.46 ±0.29) s; 40 min: (9.28 ±0.73) s; 50 min: (9.41 ± 0. 32) s] were significantly higher than in group A (P 〈0.05 ). In group C, MWT at each time point [10 min: (21.02 ±0.29) g; 20 min: (20.89 ±0.35) g; 30 min: (20.24 ±0.62) g; 40 min: (21.71±0.36) g; 50 min: (21.07 ±0.74) g], and TWL [10 min: (10.46 ±0.23) s; 20 min: (11.21 ±0.74) s; 30 min: (10.97 ±0.27) s; 40 min: (10.62 ±0.46) s; 50 min: (10.71 ± 0. 39) s ] were significantly higher than in group A (P 〈 0. 01 ). Conclusion Lacosamide administrered intraperitioneally can produce a better analgesic effect in the rat model of neuropathie pain.
