Relationships of HIF-1α,VEGF and sFlt-1 to Severe Preeclampsia

摘要 目的探讨缺氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)及可溶性血管内皮生长因子受体-1(sFlt-1)与重度子痫前期发病的关系。方法选择择期剖宫产终止妊娠的孕妇60例,其中重度子痫前期患者(研究组)30例,正常孕妇(正常对照组)30例。采用免疫组织化学SP法检测2组孕妇胎盘组织中VEGF的表达,酶联免疫吸附试验检测2组孕妇血清VEGF、sFlt-1和HIF-1α的表达水平。结果与正常对照组比较,研究组孕妇胎盘和血清VEGF表达水平均显著降低,血清HIF-1α、sFlt-1表达水平均显著升高(均P<0.01)。结论 HIF-1α可能通过VEGF、sFlt-1参与重度子痫前期的发病。 Objective To investigate the relationships of hypoxia-inducible factor-1α(HIF-1α),vascular endothelial growth factor(VEGF)and soluble fms-like tyrosine kinase-1(sFlt-1)to severe preeclampsia.Methods Sixty pregnant women scheduled for elective cesarean delivery were divided into study group(30patients with severe preeclampsia)and normal control group(30normal pregnant women).The expression of VEGF in placental tissue was measured by immunohistochemistry.The levels of HIF-1α,VEGF and sFlt-1in serum were detected by ELISA.ResultsCompared with normal control group,the expression of VEGF in placental tissue and serum decreased,but the levels of HIF-1αand sFlt-1increased in study group(P〈0.01).ConclusionHIF-1αmay be involved in the pathogenesis of severe preeclampsia through regulating the expression of VEGF and sFlt-1.
