摘要
目的寻求可靠、稳定、适宜工业化大生产的生附子中抗肿瘤成分纯化工艺。方法通过静态实验筛选出大孔树脂,并对纯化工艺条件及参数采用动态实验进行研究。以生附子总生物碱提取率为评价指标,用紫外分光光度法测定总生物碱含量。结果确定树脂类型为AB-8,上样浓度为1g/ml,上样流速为2BV/h,用2BV氨性乙醇以2BV/h流速洗脱,得生物碱平均收得率为53.6%。结论 AB-8型大孔树脂对生附子中总生物碱纯化效果较好,工艺稳定,可用于生附子中总生物碱纯化的产业化生产。
Objective To seek for the purification process of antitumor components,reliable and also stable suitable for industrialized production of Aconite roots. Methods The macroporous resin was selected through the static experiment,and the purification process conditions and parameters were studied by dynamic experiments. Take the extraction rate of total alkaloids of Aconite roots as the evaluation index,determination of total alkaloid content by Ultraviolet Spectrophotometry. Results The resin type is AB- 8,the concentration is 1g / m L and the sample current velocity is 2BV / h,the average yield( %) of the alkaloids was 53.6%. with the 2BV ammonia alcohol flow rate of 2BV / h. Conclusion AB- 8 type of macroporous resin for total Alkaloids of aconite roots is good purification effect,process stability,Which can be used for the purification of total Alkaloids of aconite roots in industrial production.
