慢性丙型病毒性肝炎患者CD4^+CD25^+ Treg细胞与自身抗体的相关性研究 被引量:7

摘要 目的探讨慢性丙型病毒性肝炎患者CD4^+CD25^+Treg细胞水平与自身抗体之间的关系及其临床意义。方法选取2013年10月至2015年2月该院收治的324例新入院且未经治疗的慢性丙型病毒性肝炎患者,利用间接免疫荧光和免疫印迹方法检测患者自身抗体谱的表达状况,同时使用流式细胞术计数患者外周血CD4^+CD25^+Treg细胞数量。结果 324例慢性丙型病毒性肝炎患者,有136例患者至少有1项自身抗体阳性,另有188例慢性丙型病毒性肝炎患者自身抗体谱表达均为阴性。自身抗体阳性组CD4^+CD25^+Treg细胞百分比明显低于自身抗体阴性组(P<0.05);同时慢性丙型肝炎患者CD4^+CD25^+Treg细胞水平随着自身抗体表达数的增加而降低,呈负相关。结论对HCV感染患者进行CD4^+CD25^+Treg细胞水平监测,能有效预判患者发生免疫耐受的可能性。 Objective To investigate the relationship and the clinical significance between CD4~+CD25~+Treg cells and autoantibody in patients with Chronic Hepatitis C.Methods Newly hospitalized and untreated 324 patients with chronic hepatitis C in our hospital were selected from October in 2013 to February in 2015.Using the methods of indirect immunofluorescence and Western blot,we observed the expression of serum autoantibody profiles in hepatitis C patients.Meanwhile,the amount of CD4~+CD25~+Treg cell in peripheral blood of patients with Chronic Hepatitis C were detected by flow cytometry.Results 136 patients with chronic hepatitis C in 324 cases had at least one positive autoantibody before treatment,none of other 188 patients had a positive autoantibody.The amount of CD4~+CD25~+Treg cell in peripheral blood of patients with Chronic Hepatitis C prompted that the percentage of CD4~+CD25~+Treg cells in the positive group was significantly lower than that in the negative group(P〈0.05).What′s more,CD4~+CD25~+Treg cells of patients decreased with the increase of the positive number of autoantibody,which was negatively correlated with each other.Conclusion Monitoring the level of CD4~+CD25~+Treg cell in patients with HCV infection could effectively predict the possibility of immune tolerance.
