甲状腺激素受体相互作用因子4基因多态性与阿尔茨海默病的相关性研究 被引量:7

摘要 目的 探索甲状腺激素受体相互作用因子4 (TRIP4)基因rs4776494多态性与阿尔茨海默病的相关性.方法 运用病例对照研究设计,收集安徽及周边地区77例拟诊为阿尔茨海默病患者和80例正常健康对照.通过聚合酶链反应(PCR)及测序的方法,比较病例组和对照组TRIP4rs4776494的等位基因及基因型频率的差异.并对每例研究对象进行简易精神状态检查表(MMSE)、临床痴呆评定量表(CDR)、痴呆伴精神行为异常(BPSD)量表检测.结果 TRIP4基因rs4776494的TT、CT和CC基因型在病例组和对照组中的频率公布[TT、CT、CC基因频率分别是AD组:57/77(74.02%)、18/77 (23.38%)、2/77(2.6%);正常组:38/80(47.5%)、39/80(48.75%)、3/80(3.75%)]均差异有统计学意义(P<0.01).病例组中TRIP4基因携带者BPSD出现率明显高于非携带者(P<0.05).结论 TRIP4(rs4776494)T型等位基因是AD的风险基因.TRIP4基因可能在一定程度上促进了患者精神症状的发生. Objective To explore the association between the common variations of thyroid hormone receptor interactor 4(TRIP4) gene rs4776494 and Alzheimer disease(AD) in Chinese people.Method A case-control study was performed,consisting of 77 patients with AD (AD group) and 80 controls (control group) who were matched at age and gender with the AD patients.PCR and sequencing were used to determine the distribution of allele and genotype frequency of TRIP4 rs4776494.Every case was subjected to minimental state examinations(MMSE),clinical dementia rating(CDR) and behavioral and psychological symptoms of dementia (BPSD).Results The TT,CT and CC gene frequencies were 57/77 (74.02%),18/77 (23.38%) and 2/77(2.6%) in AD group,and 38/80(47.5%),39/80(48.75%) and 3/80(3.75%) in control group,respectively.T allele in cases group was significantly higher than that in control group (P< 0.01).Cases with T allele were more susceptible to BPSD than those without T allele(P<0.05).Conclusion T Allele of TRIP4 rs4776494 might be a genetic risk factor of AD in Chinese population.To some degree,TRIP4 gene may be associated with mental symptoms in AD patients.
