β-连环蛋白在铜绿假单胞菌感染时的调节作用 被引量:1

摘要 目的探讨在铜绿假单胞菌感染时β-连环蛋白(β-catenin)对细菌感染引起的炎性反应和细菌清除的调节作用。方法采用Western-blot和免疫荧光检测细菌感染前后β-catenin的表达;慢病毒感染后Western-blot检测β-catenin上调效果;实时荧光定量聚合酶链反应(RT-PCR)检测促炎性因子的表达;平板计数检测荷菌量。结果在铜绿假单胞菌感染后,β-catenin在巨噬细胞和中性粒细胞中的表达呈现一过性上调后逐渐降低;β-catenin在细胞中抑制促炎因子的表达,并促进细菌清除。结论在铜绿假单胞菌感染时β-catenin抑制炎性反应并促进细菌的清除。 Objective To explore the regulating role of β-catenin in bacterial infection caused inflammation reaction and bacterial clearance after Pseudomonas aeruginosa infection.Methods Western-blot and immunofluorescence assay were used to determine the β-catenin expression in macrophage-like RAW264.7cells and neutrophils before and after Pseudomonas aeruginosa infection;the upregulating effect of β-catenin after lentivirus infection was detected by Western-blot;the pro-inflammatory cytokine expression was examined by using real-time PCR(RT-PCR);the bacterial load was assessed by using the plate count assay.Results The β-catenin protein expression in Pseudomonas aeruginosa infected macrophages and neutrophils showed the transient upregulation and then was gradually decreased.β-catenin suppressed the expression of pro-inflammatory cytokines and promoted bacterial clearance in Pseudomonas aeruginosa infected macrophages and neutrophils.Conclusion β-catenin inhibits the inflammation reaction and enhances the bacterial clearance in Pseudomonas aeruginosa infection.
