摘要
作者用~3H-胸腺嘧啶核苷掺入法筛选出的3株单克隆抗体(McAb)对L.d.前鞭毛体具有抑制作用,并以2B12-A8及2H6-E3作用最强,在补体协同作用下,其抑制率稳定在94~99.3%的优异水平。而1B1-C2加补体的抑制率波动于68~95%.用McAb 1B1-C2对L.d.同株前鞭毛体分别进行~3H-胸腺嘧啶核苷掺入法及巨噬细胞吞噬抑制试验,两法所得抑制率极为相近,分别为68~95%及61~87%。经免疫金技术超微定位证实,抑制作用最强的McAb 2H6-E3识别的抗原较广泛地存在于前鞭毛体膜外沿。此保护性McAb的发现,为黑热病免疫预防的研究提供了重要基础。
Kala-azar is increasing in incidenc insome endemic areas of China, such asprovinces of Sichuan and Gansu. Monoclo-nal antibodies nave been shown to inter-fere with the cutaneous leishmaniasisinfection in the experimental animals. Wehave now raised McAb against membraneantigen of promastigotes of L.donovanicanine isolate (the pathogen of visceralleishmaniasis).In this paper we report 3McAb which inhibit the growth of pro-mastigotes in vitro culture by incor-poration of ~3H-thymine deoxyriboside,suchas McAb 2B12-A8, 2H6-E3, in the presenceof complement, showing strong inhibitingeffects with a decrease in incorporation of^3H-TdR, 94-99.3% rate of inhibition, whereasthe inhibition rate induced by McAb 1B1-C2fluctuated in a range of 68-95%, which isidentical with the results of macrophageinfection inhibition test by the same McAb,61-87%. The results of ultrastructural locali-zation of L. donovani antigen by pro-tective McAb 2H6-E3 with immunogoldtechnique showed the godd particies, inclunps, widely distributed on the outerside of promastigotes membrane. Accordingto the high density of localized goldparticles, it is suggesed that this antigenis abundant on the plasma membrane.
