新型萘醌类衍生物对细胞衰老的抑制作用 被引量:14

Inhibitor Effects of Novel Naphthoquinone Derivative Preventing Cellular Senescence on Mouse Embryonic Fibroblast

摘要 通过对萘醌类化合物的化学修饰,研究2-取代5,8-二甲氧基-1,4-萘醌类衍生物对鼠胚胎成纤维细胞(MEF)衰老的抑制作用。应用噻唑蓝(MTT)、流式细胞术(FCM)、SA-β-gal染色等方法检测多种萘醌衍生物对细胞毒性、细胞内活性氧(ROS)水平和细胞衰老的作用,实验结果显示:2-苄硫基-5,8-二甲氧基-1,4-萘醌在降低其细胞毒性作用的同时,具有清除细胞内活性氧从而抑制细胞衰老的作用。 To study inhibitory effect of novel naphthoquinone derivative on cellular senescence,the chemical structure of naphthoquinone compounds was modified to study the inhibitory effect of 2-substitude 5,8-dimethoxy-1,4-naphthoquinone derivatives on mouse embryonic fibroblast(MEF) senescence. The cytotoxicity of a variety of naphthoquinone derivatives were tested by MTT assay,and were detected the intracellular ROS level and cellular senescence situation by flow cytometry (FCM) and SA-β-gal staining. All of results indicated that 2-benzylthio-5,8-dimethoxy-1,4naphthoquinone not only had the lower cytotoxicity,but also had the role to prevent cellular senescence by intracellular ROS.
