摘要 激光共聚焦显微镜(共聚焦显微镜)是在生物学研究中最通用的荧光显微镜工具之一。它的广泛流行是因为其优越的光学切片和3维(3D)重建功能。不同于普通显微镜观测厚样本时由于被聚焦外光线干扰导致模糊图像,激光共聚焦显微镜可以从比较厚的生物样本在短期内获得清晰的光学切片。激光共聚焦显微镜所得到的一幅完全对齐的3D图像系列允许对样本在虚拟空间的任意角度观测。该技术越来越成为现代生命科学研究中必不可少的一项研究工具。本文的目的是为研究生和青年研究人员提供一些使用激光共聚焦显微镜的基础知识并对它的应用做简单的介绍。 Confocal Laser Scanning Microscopy(CLSM)is one of the most versatile fluorescence microscopy tools available in biological researches.Its popularity is mostly because of its remarkable optical sectioning and 3-Dimension(3-D)reconstruction capabilities.A CLSM could obtain sharp optical sections without out-of-focus blurs from a relatively thick biological specimen in a short period of time.A perfectly registered 3-D image stack from a CLSM allows extraordinary virtual visualization of the specimen.This technology review paper is intended for graduate students as well as young researchers to have some basic knowledge of the technology and intended to provide some guidelines for proficient usage of the equipment.
