钩端螺旋体冷冻蚀刻复型的超微结构研究

摘要 作者应用冷冻蚀刻复型技术研究3株钩用螺旋体的超微结构,发现:①钩体在冷冻蚀刻复型下的形态与超薄切片及负染的观察结果基本一致,不同种属与毒力的钩体其冷冻蚀刻复型的超微结构无明显差异;②钩体外膜蛋白颗粒呈球形,其分布明显不对称。在外膜的EF面,蛋白颗粒密集分布,其密度为776~2303个/μm^2,直径为14.25±2.25nm,而PF面则甚少,呈平滑状;③轴丝位于圆柱形菌体与外膜之间,直径20nm,外周有一7nm鞘状结构包绕;④钩体细胞质中有核糖体、核质及包涵体等亚细胞结构存在。本研究结果表明:冷冻蚀刻复型技术较其他方法能更清晰地显示接近生活状态的钩体的超微结构。 The ultrastructure of three strains ofleptospires,i. e. L. interrogans serovar Laistrain 017, L. biflexa serovar patoc strainPaoc Ⅰ and L. illini strain 3055,were stud-ied with the technique of freeze-etchingreplica. The results showed that (1) theultrastructure of lepospires after freeze-etching are similar to that observed bymeans of negative staining and ultrasec-tioning.There are no dramatic differencesamong strains of leptospires studied. (2)Globular particles exposed on the resultingtwo inner membrane faces are asymmet-rically distributed. Large areas on EF facestudied with numerous globular particles,whereas the PF face had few of them. Thedensity of globular particles varies in differ-ent parts of leptospires,from 776/μm^2 to2303/μm^2.The globular particles, 14.25±2.25 nm in diameter,represent membraneproteins in outer envelope of leptospires.(3) Axial filament, 20 nm in diameter,were seen closely surrounded by a 7 nmsheath-like structure. (4) There are subcel-lular structures in cytoplasm of leptospires.They appear to be ribosomes, chromatinsand inclusions.
