摘要
利用四种随机引物对采自青岛附近的三种海洋盾纤目纤毛虫 :弗州拟尾丝虫(Parauronemavirginiaum ,Thompson)、中国后阿脑虫 (Metanophryssinensis ,Song&Wilbert)及相似后阿脑虫 (Metanophryssimiles ,Songetal)进行了RAPD分析 ,对这三种亲缘关系密切的形态种做了分子生物学水平的廓清 ;同时利用筛选出的三种限制性内切酶 ,对上述纤毛虫 16s小亚基单位、2 3s大亚基单位核糖体DNA区间包括其内转录间隔序列的基因片段进行切割 ,结果表明所得限制性片段多样性图谱在重复实验中表现出了良好的稳定性。本工作显示 ,两种方法所得图谱均可将 3种纤毛虫明显区分。文中还对上述技术在本领域的适用性。
Three closely-related scuticociliates were investigated using random amplified polymorphism DNA fingerprinting (RAPD) and restricted fragment length polymorphism (RFLP). Genome of 3 scuticociliates were amplified with 4 random primers, and polymerase chain reaction (PCR) was perfomed to amplified the 16s~23s rDNA segment, followed by ristriction digestion to yield restriction fragment length polymorphisms. Special bands of the RAPD products of each species and distinctly different restriction disgestion haplotypes distinguished the three species. The present paper gives the discussion about the applicability, reaction conditions and systems of the two methods.
