摘要
以大樱桃8个品种为研究对象,利用改进CTAB法进行DNA提取,使用10碱基随机引物,通过PCR进行RAPD(Random Amplified Polymorphic DNA)扩增,建立了鉴定大樱桃品种的分子鉴定图谱。结果表明:较高质量的DNA和理想的RAPD反应体系能够保证RAPD技术具有较好的稳定性;筛选出的不同引物的RAPD扩增谱带组合可用于大樱桃栽培品种的分子鉴定,具有在生产应用的潜在性。
An improved DNA extraction protocol of CTAB was developed for 8 varieties of sweet cherry.Based on the results of various assays, the reliable RAPD protocol for sweet cherry was screened.And the molecular genetics marks which were used to distinguish varieties of sweet cherry were established.The results showed that high-quality total DNA and RAPD marker technique could be a reliable screen for identifying various varieties of sweet cherry.The combined RAPD fingerprints of sweet cherry which were derived from primers with diverse numbers of nucleotides in length were different in band numbers, polymorphism and band clarity and shed light into the potential practice in identification of distinct sweet cherry varieties in field.
