摘要
采用新的化学方法将3.0μm无孔聚甲基丙烯酸环氧丙酯树脂改性为一种新型的两性离子交换色谱填料。本文主要考察了该填料对标准蛋白的分离性能,流速等对蛋白质保留的影响。结果表明,本文合成的两性离子交换色谱填料具有优异的色谱性能,能同时达到对酸性和碱性蛋白的分离纯化。将其应用于快速纯化重组人粒细胞-巨嗤细胞集落刺激因子(rhG-CSF),效果较好。
3.0 μm Non-porous poly(glyciddylmethacrylate-co-ethylenedimethacrylate) particles were modified to be a zwitterionic exchange(ZIC)stationary phase for HPLC by new chemical method. The synthesis of zwitterionic stationary phases has advantages of biopolymer separation, and was capable of separating acidic and basic proteins simultaneously. The zwitterionic resin was used for purification of rh G-CSF, the satisfactory results were obtained.
