摘要
目的探讨肢体缺血后适应(LIPost C)对脑缺血-再灌注后小鼠血-脑屏障(BBB)损伤的保护作用及紧密连接蛋白claudin-5、occludin的表达变化。方法将雄性CD1小鼠90只随机分成:假手术组(Sham组)、缺血-再灌注组(I/R组)、肢体缺血后适应组(LIPost C组),采用改良线栓法制备大脑中动脉缺血-再灌注模型。术后24 h,分别进行神经功能缺损评分、脑含水量、脑梗死体积、BBB渗透性检测,采用Western blot和实时荧光定量PCR(RT-q PCR)检测claudin-5、occludin的表达。结果与I/R组相比,LIPost C组神经功能缺失评分提高,梗死体积减小,脑水肿减轻、BBB渗透性下降,Claudin-5、occludin的表达上调,差异均具有统计学意义(均P<0.05)。结论肢体LIPost C对脑缺血-再灌注后小鼠BBB具有保护作用,其保护机制与上调claudin-5、occludin的表达密切相关,为其临床应用提供理论基础。
Objective To investigate the protective effect of Limb ischemic postconditioning( LIPost C) on blood brain barrier( BBB) and explore the influence on the the expression of tight junction protein claudin-5,occludin in cerebral ischemia-reperfusion injury of mice. Methods Ninety male CD1 mice were divided into three groups randomly: Sham-operated group( sham group),ischemia-reperfusion group( I / R group) and LIPost C group. Mouse middle cerebral artery cerebral ischemia-reperfusion model was established by modified-occlusion method.Neurological deficit scores,brain water content,infarct volume,BBB dysfunction were measured at 24 h after reperfusion. Western blot and RT-q PCR were used to analyze the expressions of claudin-5,occludin. Results Compared with I / R group,neurological deficits scores were significantly decreased,brain edema was relieved,infarct volume was reduced,and the expression of claudin-5,occludin up-regulated,the differences were statistically significant( all P〈0. 05). Conclusion LIPost C can protect the BBB of cerebral ischemia-reperfusion injury mice by increasing the expression of claudin-5,occludin,and that will provide a theoretical basis for its clinical application.
