摘要
为筛选水黄皮内生菌中对水稻害虫有毒杀作用的菌株,从健康水黄皮植物的根、茎、叶和种子中进行菌株分离,并对分离出的菌株的形态、培养特征、生理生化及其他生物学特性进行研究;通过16S r DNA及ITS序列的系统发育学对其分析和鉴定。结果表明:用玻片浸渍法和喷雾法筛选出杀虫活性物质菌株,共获得细菌88株、放线菌12株、真菌3株。经初筛及复筛,内生菌麦角菌[Clavicepspurpurea(St F1)]和蜡样芽胞杆菌[Bacillus cereus(Se B3)]的发酵液粗提物分别对稻飞虱(Rice planthopper)、稻纵卷叶螟(Cnaphalocrocis medinalis Guenee)致死率达85%以上。水黄皮内生菌株Se B3和St F1的粗提代谢物对稻纵卷叶螟和稻飞虱有明显地毒杀作用,为开发新的生物源农药提供了可能的生物质。
This study was performed to investigate endophyte species of Pongamia pinnata and to screen strains with insecticidal activity against rice insect. The endophytes were isolated from roots,stems,leaves and seeds of health P. pinnata by grinding separation method and identified by a dual approach of morphological and physiological observation and 16 S r DNA gene(for bacteria) and ITS sequence(for fungi) based molecular identification. The endophytes were inoculated in liquid media for fermentation and the crude extracts were used to test insecticidal activities by slide disc immersion and nebulization methods.The result showed that endophytic species were classified into 9 genera of bacteria,3 actinomycetes and 3 fungi were isolated.The strain Bacillus cereusi(Se B3) and Claviceps(St F1) showed more than 85% of mortality against rice planthopper and cnaphalocrocis medinalis Guenee. Two strains of Se B3 and St F1 associated within the P. pinnata had significant insecticidal activity to rice insects,which may provide a new biological resource to explore new microbial insecticide.
