DHA联合葫芦素B对人胃癌BGC-823细胞增殖及凋亡的影响 被引量:1

摘要 目的:研究二十二碳六烯酸(docosahexaenoicacid,DHA)联合葫芦素B(cucurbitacin B)体外对人胃癌BGC-823细胞增殖及凋亡的影响。方法:利用NADH酶染色、吉姆萨染色显微镜下观察人胃癌BGC-823细胞经DHA联合葫芦素B作用后细胞形态学上的变化。采用MTT法、流式细胞仪检测细胞增殖、周期及凋亡的情况。结果:根据MTT法DHA联合葫芦素B作用于胃癌细胞比分别利用DHA、葫芦素B直接作用细胞增殖抑制更为明显,更有效降低细胞的存活率,且在相同时间随着药物浓度的提高,作用效果也随之提高。NADH酶染色法、吉姆萨染色法在显微镜下明显可见细胞凋亡现象。流式细胞仪检测肿瘤细胞,药物联合干预G0/G1期细胞明显增多,G2/M期和S期细胞明显减少。结论:DHA联合葫芦素B可抑制人胃癌BGC-823细胞增殖、诱导细胞凋亡,其联合具有协同作用。 Objective: To investigate the combined effects of docosahexaenoic acid( DHA) and Cucurbitacin B on cells proliferation and apoptosis of human gastric cancer BGC- 823 cells. Methods: BGC- 823 cells were stained with NADH enzyme and Giemsa to observe their morphological changes under the microscope before and after treated with DHA and Cucurbitacin B. MTT assay and flow cytometry were used to measure proliferation,cycle and apoptosis of BGC- 823 cells respectively. Results: According to the MTT assay,compared with the effects of DHA and Cucurbitacin B respectively,the cell viability was reduced more effectively with the combined treatment on cell proliferation inhibition of gastric cancer cells,and the reduction effect improved as the drug concentration increased. With NADH enzyme and Giemsa staining,apoptosis phenomenon could be observed visibly by microscope. When flow cytometry was employed on the tumor cells with drugs joint intervention,G0/ G1 cells increased significantly while G2/ M and S phase cells decreased significantly. Conclusion: The combined treatment of DHA and Cucurbitacin B can inhibit the growth and induce the apoptosis of human gastric cancer BGC- 823 cells,and has synergistic effect.
