摘要
目的探讨白藜芦醇与涎腺腺样囊性癌细胞系ACC-M增殖及凋亡的相关性。方法使用终浓度各为12.5、25、50、100和200μmol/L的白藜芦醇处理ACC-M24、48、72 h,并用MTT比色、倒置显微镜观察、光镜观察、流式细胞仪检测并观察ACC-M细胞增殖及凋亡状况。结果白藜芦醇对ACC-M生长成时间-浓度依赖性抑制,最大抑制率可达92.6%。白藜芦醇对ACC-M凋亡有诱导作用,凋亡率与经白藜芦醇作用时间及作用浓度呈正相关(P<0.05)。白藜芦醇可造成ACC-M坏死。结论白藜芦醇通过诱导涎腺腺样囊性癌细胞系ACC-M凋亡及坏死,抑制ACC-M增殖。
Objective To observe the effect of resveratrol on human salivary adenoid cystic carcinoma cell line-ACC-Min vitro. Methods The ACC-M cells were treated with resveratrol at 12. 5,25,50,100,200μmol / L for 24 h,48h,72 h,respectively. The proliferation and apoptosis of ACC-M cells were observed by MTT assay,inverted microscopy,light microscopy,flow cytometry. Results Resveratrol inhibited the proliferation of ACC-M cells in a dose and time dependent manner. The highest inhibition rate was 92. 6%. Resveratrol induced apoptosis and necrosis of ACC-M cells,and the apoptotic rate was positively correlated to the action time and action concentration of resveratrol( P〈0. 05). Conclusion Resveratrol can inhibit the proliferation of ACC-M cells by inducing apoptosis and / or necrosis of the cells.
