鸡白痢沙门菌中三型分泌系统-2效应蛋白基因分布研究 被引量:1

Distribution of Type Ⅲ Secretion System-2 Effector Genes Among Salmonella Pullorum Isolates

摘要 为了解鸡白痢沙门菌中三型分泌系统-2(T3SS2)效应蛋白基因的分布情况。本研究采用PCR方法检测了在1962~2013年间从我国分离的237株鸡白痢沙门菌中25个T3SS2效应蛋白基因的分布特征;同时,通过序列比对分析了这25个基因在18株不同血清型沙门菌中的流行特征,它们的全基因组序列来自GenBank。PCR结果显示,在这237株菌株中,cigR、pipB、pipB2、slrP、sopD、sopD2、spiC、sseF、sseG、sseJ、sseK1、sseL、steA、steB、steC和steD均存在,gogB和sspH1均缺失,其它基因分布水平在5.9%~99.6%之间。序列比对分析结果显示,有15个基因在这18株菌株中均存在,sifB、sseK1、sspH2和steC在一些菌株中是假基因。结果表明,gtgE、sseI和sseK2在这237株菌株中分布具有一定的年代规律性;大部分被检测基因在鸡白痢沙门菌中具有广泛分布;一些基因在微进化过程中形成了假基因。 To investigate the distribution of type Ⅲ secretion system-2(T3SS2)effector genes among Salmonella Pullorum isolates. We determined the distribution of 25 T3SS2 effector genes among 237 Salmonella Pullorum isolates in China from 1962 to 2013 by PCR. Meanwhile,we also analyzed the epidemic characteristics of these genes in 18 different serovars Sallmonella strains whose genome sequences were downloaded from Gen Bank. PCR determination of25 T3SS2 effector genes demonstrated the presence of cig R,pip B,pip B2,slr P,sop D,sop D2,spi C,sse F,sse G,sse J,sse K1,sse L,ste A,ste B,ste C and ste D genes and the absence of gog B and ssp H1 genes in these isolates,other genes were variably distributed with frequency values ranging from 5.9% to 99.6%. Sequence analysis revealed that 15 genes of cig R,pip B,pip B2,sif A,sif B,slr P,sop D,sop D2,spi C,sse F,sse G,sse L,ste A,ste C and ste D genes exist in all of the 18 different serovars strains;sif B,sse K1,ssp H2 and ste C exist as pseudogenes in some of the 18 strains. Our results indicated that gtg E,sse I and sse K2 genes have chronological change of prevalence among the 237 Salmonella Pullorum isolates,most of the 25T3SS2 effector genes have extensive distribution,some genes are pseudogenes during microevolution process.
