摘要
目的:建立简单、快速、灵敏的测定花生壳中木犀草素含量的的液相色谱-单级质谱联用(LC-MS)方法。方法:采用Kromasil C18(4.6mm×50mm,5μm)色谱柱;流动相:甲醇-0.2%甲酸(体积比50∶50);流速为0.35m L·min-1;柱温为室温;质谱条件:电喷雾离子源(ESI),选择性离子检测(SIM),正离子模式。结果:木犀草素在50~2000ng·m L-1浓度范围内,线性关系良好(r2=0.9958);方法的精密度、重现性和稳定性良好;平均回收率为97.70%。采用该方法测定的花生壳中木犀草素的平均含量为0.21%。结论:该法专属性强、快速、灵敏,可用于花生壳中木犀草素的含量测定。
Objective: To develop a simple, quick, sensitive LC-MS method for the determination of luteolin in peanut shell. Method: The analysis was performed on a Kromasil C18 column (4.6mm×50mm, 5μm) with the mo- bile phase of methanol- water (containing 0.2% formic acid)(v/v=50:50) at a flow rate of 0.35 mL.min-1. The temperature of column was room temperature. The LC-MS system was operated by using an electrospray ionization (ESI) probe in the positive ion mode. Scan mode was in selected ion monitoring (SIM). Result: The calibration curve of luteolin was linear in the range of 50-2000ng.mL-1 (r2=0.9958). The precision and repeatability and sta- bility were in line with the sample measurement requirements. The mean recoveries were 97.70%. The average content of luteolin in peanut shell was 0.21%. Conclusion: The method was proved to be selective, quick and sen- sitive and have been successfully applied for determination of luteolin in peanut shell.
