摘要
目的:观察有氧运动对非酒精性脂肪肝大鼠脂联素、AdipoR2及PPARα的影响,并探讨其可能机制。方法:30只SD雄性大鼠随机分为正常组(N组)、模型组(C组)和运动组(E组),N组喂食基础饲料,C组和E组均喂食高脂饲料,E组大鼠进行12周游泳训练。12周末取材观察肝组织切片,并对其炎症活动度评分,检测比较各组大鼠肝组织中TC、TG、FFA含量和血清脂联素、TC、TG、ALT、AST、FFA含量的变化;肝组织AdipoR2、PPARα蛋白表达水平。结果:(1)肝组织切片显示,N组大鼠肝组织学特征正常,没有肝细胞脂肪变性及炎症浸润;C组大鼠肝组织可见弥漫性肝细胞脂肪变性,肝板排列不整齐,肝小叶内炎症细胞浸润,出现汇管区炎症。E组大鼠肝细胞形态基本正常,肝板排列较整齐,肝小叶内及汇管区少见炎症细胞浸润;C组大鼠肝脏炎症活动度评分显著高于N组和E组。(2)C组大鼠肝组织TC、TG、FFA水平显著高于N组和E组;C组大鼠血清TC、TG、FFA、ALT、AST水平均显著高于N组和E组。(3)C组大鼠肝组织AdipoR2、PPARα蛋白表达水平显著低于N组和E组。结论:有氧运动可以通过提高非酒精性脂肪肝大鼠血清脂联素水平同时激活肝组织AdipoR2、PPARα蛋白表达发挥防治非酒精性脂肪肝的作用。
Objective: This experiment was to investigate the effect of exercise on adiponectin, AdipoR2 and PPARαprotein activity in Liver,and study the mechanism.Methods: 30 male SD rats were randomly assigned to three groups: control group( N); high fat diet group( C); exercise and fed by high fat diet group( E). The group E underwent swimming for 12 weeks. At the end of experiment,The expression of AdipoR2 and PPARα in liver tissue were tested. The content of TC,TG,FFA in liver tissue and TG,TC,FFA,ALT,AST in serum were detected. Results:( 1) The hepatic sections in group C showed severe macrovesicular steatosis,and the steatosis severity. The steatosis in group E was improved obviously compared with that in group C( 2)The serum TG、TC、ALT、AST of C group was higher than that of other groups; The content of TC,TG,FFA in liver tissues in C group was the highest;( 3) The expression of AdipoR2 and PPARαin liver of N and E groups were significantly higher than that of C group. Conclusion: It suggested that aerobic exercise is likely to one of the factor by significantly increase AdipoR2 and PPARαin liver tissue anti-nonalcoholic fatty liver.
