用SDS—聚丙烯酰胺凝胶电泳分析麻风菌抗原成分初探

摘要 用SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)法对犰狳风菌(ML)和鸟型、海鱼、耻垢、瘰疠及BCG等分枝菌的超声物上清蛋白抗原做了初步分析。将ML及另5种分枝菌的超声物上清液在15%丙烯酰胺的分离胶上电泳后,经考马斯亮蓝染色得到各菌的抗原谱带图。结果显示:ML及另5种分枝菌的抗原谱带均不完全相同。ML的总带数仅及其它分枝菌的一半,其所显示的20条抗原带中主要成分为14.5、16.5、20、22、25、40、52、60、65和72KD蛋白,鸟型的50KD带,耻垢的31KD带和BCG的24.5KD带较明显。这提示分枝菌的抗原分析在菌种鉴别和鉴定、寻找血清诊断法中的有效吸收剂及有免疫作用的抗原等方面,均有一定的实际意义和潜力。文中对实验技术做了讨论。 A preliminary study has been conducted on anti- genic analysis of M. leprae (ML) derived from infected armadillos, M. avium, M. marinum, M. smegmatis, M, scrofulaceutn and BCG. Firstly, the supernatants of 6 bacterial sonic extracts were fractionated by SDS - PAGE using 15% polyacrylamide gel. Subsequently. the proteins in the gel were stained with Coomassie brilliant blue and the separated protein bands from mycobacteria were obtained. The results indicated that the spectra of the antigen bands from 6 mycobacteria weree different. The number of antigen bands in ML is only half of the other 5 species of mycobacteria, displaying 20 protein bands in which 14. 5KD, 16. 5KD, 20KD, 22KD, 25KD, 40KD, 52KD, 60KD and 72KD bands are the major components.50KD prortein band in M.avium., 31KD band in M.smegmtis and 24. 5KD band in BCG are more marked. The authors think that the spectrum of antigen bands has an important and potential signifficance for distinguishing and identifying species of mycobacteria and preparing effective absorbent reagents for serodiagnos- tic tests, and even it favoured finding new antigens which could play an important role in immunologic reations for leprosy. Relevant techniques for sensitivity, stability and reproducity of SDS--PAGE were discussed in detail.
