便塞通合剂对便秘模型大鼠PGP9.5及SS在结肠组织中表达的影响 被引量:3

Effection of Biansetong Mixture on PGP9.5 and Somatostatin in rats with Constipation

摘要 [目的]观察便塞通合剂对实验性便秘大鼠PGP9.5、SS的影响及其效果差异。[方法]将80只SD大鼠随机分为空白对照组、模型组、麻仁丸组、便塞通合剂组,每组20只,采用改良复方地芬诺酯法制作大鼠实验性便秘模型,除空白对照组和模型组不进行药物治疗外,麻仁丸组和便塞通合剂组分别给予麻仁丸0.3g·kg-1·d-1灌胃,便塞通合剂7.5g·kg-1·d-1灌胃。14d后处死全部大鼠,观察大鼠结肠组织病理学改变,采用免疫组织化学法检测结肠组织中PGP9.5、SS的表达水平。[结果]HE染色空白对照组大鼠结肠黏膜正常,模型组大鼠结肠黏膜有炎性细胞浸润,肌间神经丛神经细胞减少,可见空泡变性。通塞便合剂治疗后,近端结肠黏膜未见炎性细胞浸润,肌间神经纤维未见空泡变性。与对照组比较,模型组SS表达显著增加(P<0.05),PGP9.5表达显著减少(P<0.05);麻仁丸组和便塞通组均表现为下调SS的高表达状态,明显低于模型组(P<0.05),上调PGP9.5,明显高于模型组(P<0.05);而便塞通合剂组对其表达影响明显优于麻仁丸组(P<0.05)。[结论]便塞通合剂对便秘大鼠有较好的疗效,主要表现在控制黏膜炎症反应、降低组织中SS的表达水平,上调PGP9.5表达。 [Objective]To observe the effect of Biansetong Mixture on Protein gene product 9.5 (PGP 9.5) and Somatostatin in rats with constipation and investigate the differential therapeutic effect. [Methods] Eighty SD rats were randomly divided into control group, model group9 MaRen Pill group and Biansetong Mixture group. Constipation rat model were made by irrigating with Diphenoxylate. The rats of control group and model group were fed without any drug. All rats were killed after 14 days. The pathology chan- ges were observed. The expression of Protein gene product 9.5 (PGP 9.5)and Somatostatin(SS)in colon tissue was detected by immunohistochemistry. [Results] The colon tissues in control group is normal. In model group, inflammatory cell infiltration were observed in colon tissues. Vacuolar degeneration was ob- served in myenteric nerve plexus and neural cells decreased. Biansetong mixture decreased inflammatory cell infiltration and vacuolar degeneration in colon tissues. Compared to control group,the expression of SS in SD rats model group was significantly increased(P〈0.05) ,and the expression of PGP 9.5 was evidently lower(P〈0.05), the expression of SS MaRen Pill group and Biansetong Mixture group was lower than model group(P〈0. 05) ,and the expression of PGPg. 5 was obviously increased(P〈0.05). In addition,the regulation of Biansetong Mixture was better than MaRen Pill, and the difference was significant (P〈0.05). [Conclusion]Biansetong Mixture has a good effect on rat model with constipation,mainly in relieving inflammatory reaction and reducing the expression of SS and increasing the expression of PGP9.5.
