The effect of metformin on proliferation and apoptosis of A549 lung cancer cells

摘要 目的研究二甲双胍(MET)体外对人肺癌A549细胞增殖和凋亡的影响。方法采用MTT法测定MET不同浓度、不同作用时间对A549细胞增殖的抑制作用;用流式细胞术检测A549细胞的凋亡;Western blot检测细胞周期蛋白Cyclin D1表达的变化;ROS荧光探针-DHE测定A549细胞中活性氧自由基(ROS)的含量。结果 MET具有抑制人肺癌细胞A549细胞增殖的作用,呈剂量和时间依赖性;MET促进A549细胞的凋亡;MET诱导A549细胞中ROS的生成;MET降低细胞周期蛋白的表达。结论 MET抑制人肺癌细胞增殖,诱导细胞凋亡,其机制可能与降低Cyclin D1的表达和诱发ROS的生成有关。 Objective To study the inhibitory effect of metformin(MET)on proliferation and apoptosis of human lungcancer A549 cells. Methods The inhibitory effect of MET on the proliferation of A549 cells was determined by MTT method.The apoptosis of A549 cells was detected by flow cytometry. The expression of cyclin D1 in A549 cells was examined byWestern blot. The reactive oxygen species(ROS)content was measured by DHE fluorescence probe. Results MET couldinhibit the proliferation of A549 cells in a dose and time dependent manner. MET could promote apoptosis of A549 cells. Inaddition, MET suppressed the expression of cell cycle protein cyclin D1 and induced the production of ROS in A549 cells.Conclusion MET can inhibit the proliferation of human lung cancer A549 cells and induce cell apoptosis. Meanwhile, METhas the ability to reducee the expression of cyclin D1 and induce ROS generation, which may be involved in the mechanism ofinhibiting lung cancer cells proliferation.
