摘要
目的研究小鼠卵母细胞减数分裂恢复过程中Akt1和Akt2在从GV期向GVBD期转化期间亚细胞定位的特点。方法采用间接免疫分析技术观察Akt1和Akt2在小鼠卵母细胞GV期和GVBD期的定位;利用显微注射实验技术将Akt1和Akt2抗体分别注射入卵细胞,在不同时间点观察GVBD的发生的数量。结果 Akt1在小鼠卵母细胞GV期均匀的分布于细胞质,而在GVBD期则分布于细胞膜。Akt2在GV期和GVBD期分布于整个细胞,但是GV期以细胞质更为密集,而在GVBD期以细胞核附近更为密集。结论 Akt1可能参与小鼠卵母细胞减数分裂恢复的调控。
Objective To study the subcellular localization of Akt1 and Akt2 in the transition from GV stage to GVBD stage during meiotic resumption of mouse oocytes. Methods The localizations of Akt1 and Akt2 in GV-oocytes and G1 fertilized eggs were observed and compared by indirect immunofluorescence analysis. GVBD were observed in different time points after GV oocytes microinjected with Akt1 and/or Akt2 antiboodies. Results Akt1 was mainly distributed in cytoplasma of GV-oocytes and in cytomembrane of GVBD oocytes. However, Akt2 was distributed in whole cells in GV and GVBD, mainly in cytoplasma of GV stage and nuclear of GVBD. Conclusion Akt1 may involved in the regulation of the meiotic resumption of oocytes.
