摘要 目的:建立HPLC法测定4种牡荆属植物(黄荆、牡荆、单叶蔓荆和蔓荆)果实中黄荆子素(VNL)的含量,比较黄荆子素的含量异同。方法:采用Zorbax Extend C18色谱柱(250 mm×4.6 mm,5μm),Extend-C18保护柱(12.5 mm×4.6mm,5μm);以乙腈-0.1%甲酸溶液为流动相,梯度洗脱,0~18 min 14.4%乙腈,18~30 min 14.4%~17%乙腈,30~45min 17%乙腈;流速1.0ml/min;检测波长254nm;柱温30℃。结果:黄荆子素在14.2~455.0μg/ml范围内呈良好的线性关系,平均加样回收率为96.99%。黄荆子素的含量在牡荆属4种植物中存在差异,其中以黄荆子和蔓荆子中含量较高。结论:该方法准确、快速,操作简便,精密度高,重复性好,可用于4种牡荆属植物中黄荆子素的含量测定。 Objective: To establish a HPLC method for the determination and comparison of the content of vitexdoin (VNL) in the fruits of four related Vitex species such as V. negundo L. , V. negundo L. var. cannabifolia (Sieb. et Zucc. ) Hand.-Mazz. , V. trifolia L. var. simplicifolia Cham. and V. trifolia L.. Methods: Analyses were performed on Zorbax Extend Cls analytical column (250 mm×4.6 ram, 5 μm) and Extend-C18 guard column (12.5 min×4.6 ram, 5 μm). Mobile phase was composed of acetonitrile-0.1% formic acid in gradient elution at a flow rate of 1.0 ml/min: 0-18 min 14.4% acetoni- trile, 18-30 min 14.4%-17% acetonitrile, 30-45 min 17% acetonitrile. Detection wavelength was 254 nm and column tempera- ture was 30 ℃. Results: VNL displayed good linearity within the ranges of 14.2-455.0 μg/ml and average additional recovery rate was 96.99%. VNL content in the fruits of the four species varied considerably,with VNL contents in V. negundo L. and V. trifolia L. being higher than those in the other two species. Conclusion: The HPLC method proves to be accurate, simple, rapid, precise and reproducible. It could be used for content determination of VNL in the fruits of the four Vitex species. ]
