荧光-磁共振双功能纳米探针FITC-CS-LA@SPION的制备及体外MRI成像 被引量:1

Preparation and in vitro MR imaging of bifunctional fluorescence and magnetic resonance nanoprobe FITC-CS-LA@SPION

摘要 [目的]制备肝细胞表面去唾液酸糖蛋白受体(ASGP-R)特异性荧光-磁共振双功能纳米探针(FITC-CS-LA@SPION),探讨其与体外肝细胞靶向结合情况及在MRI显像中的价值.[方法]利用异硫氰酸荧光素(FITC)标记壳聚糖(CS)与乳糖酸(LA)修饰的超顺磁性氧化铁纳米颗粒(SPION)合成FITC-CS-LA@SPION,采用荧光显微镜和流式细胞仪检测FITC-CS-LA@SPION与肝细胞体外结合情况,通过体外MRI成像观察细胞数量与MRI信号变化的关系.[结果]流式细胞仪检测结果表明,FITC-CS-LA@SPION与肝细胞有较强的结合能力;荧光显微镜观察结果显示,FITC-CS-LA@SPION特异性分布于肝细胞周缘,而对照组FITC-SPION仅见少量荧光分布;体外MRI成像结果显示,T2WI信号随肝细胞数量的增加而增强.[结论]FITC-CS-LA@SPION对肝细胞具有较强的特异性结合力,且具有较好的MRI增强效果. OBJECTIVE To prepare the specific bifunctional fluorescent and magnetic resonance nanoprobe(FITC-CS-LA@SPION)of asialoglycoprotein receptor(ASGP-R)on the surface of liver cells in order to investigate the hepatocytes targeting combination and value of MRI imaging in vitro.METHODS Using the Chitosan(CS)labeled by fluorescein isothiocyanate(FITC)and superparamagnetic iron oxide nanoparticles(SPION)modified by lactobionic acid(LA),the FITC-CS-LA@SPION was synthesized,and the situation of FITC-CS-LA@SPION combining with hepatocytes in vitro was detected by fluorescence microscope and flow cytometry,and the relationship between the number of hepatocytes and MRI signal changes was observed by MR imaging.RESULTS The results detected by flow cytometry showed that there was a strong binding capacity between FITC-CS-LA@SPION and hepatocytes,and by fluorescence microscopy observed that FITC-CS-LA@SPION expressed specifically in hepatocytes periphery and a small amount of fluorescence distribution in the control group,and by MRI imaging in vitro showed that the T2 WI signal increased with the increase of number of hepatocytes.CONCLUSIONFITC-CS-LA@SPION has a strong specific binding capacity to hepatocytes and good MRI enhancement effect.
