摘要
目的了解利福平耐药实时荧光定量核酸扩增检测技术(Xpert MTB/RIF)用于福建省耐多药结核病(MDRTB)利福平耐药检测的可行性。方法对来源于福建省结核病耐药性监测30个监测点纳入的79株耐多药和40株全敏感结核分枝杆菌分离菌株进行Xpert MTB/RIF检测,同时用PCR测序验证是否存在利福平耐药位点突变。结果119株菌Xpert MTB/RIF检测结果均为结核分枝杆菌。以比例法药物敏感试验结果作为对照,Xpert MTB/RIF检测MDR结核分枝杆菌利福平耐药的敏感度为97.47%(77/79),特异度为100.00%(40/40),Xpert MTB/RIF检测结果与PCR测序结果的一致性达到100%。结论 Xpert MTB/RIF方法用于耐多药结核分枝杆菌利福平耐药检测的意义较大,可用于福建省MDR-TB的快速筛查。
Objective To understand the feasibility of Xpert MTB / RIF to detect the resistance to RIF of MDR- TB in Fujian province. Methods 79 MDR isolates collected from 30 drug resistant survey sites and 40 pan- sensitive strains were detected by Xpert MTB / RIF,and the mutation of RIF was verified by PCR. Results The detection results of 119 strains by Xpert MTB / RIF all were Mycobacterium tuberculosis. Compared with the results of the proportion of drug sensitive test,the sensitivity of RIF resistance detected by Xpert MTB / RIF was 97. 47%( 77 /79),the specificity was 100. 00%( 40 /40). And the consistency rate of RIF resistance by Xpert MTB / RIF was 100% compared to the result of PCR sequenced. Conclusion It had great significance to diagnose RIF of MDR- TB by Xpert MTB / RIF,and it could help to fast detection of MDR- TB in Fujian province.
