姜黄素对心力衰竭大鼠心脏AT_1受体表达的影响 被引量:10

Effect of curcumin on AT_1 receptor expression in rat hearts with heart failure

摘要 目的研究姜黄素对大鼠慢性心力衰竭心脏血管紧张素Ⅰ型受体(angiotensin Ⅱ type 1 receptor,AT1R)的表达作用。方法结扎大鼠左冠状动脉前降支制备心肌梗死后心衰模型,32只Wistar大鼠随机分为假手术组,假手术姜黄素处理组,心衰对照组及心衰姜黄素处理组。超声心动图检测各组大鼠心功能,放射免疫法测定血浆肾素(plasma renin actvity,PRA)及血管紧张素Ⅱ(angiotensionⅡ,AngⅡ)水平,qRT-PCR检测各组大鼠心脏AT1受体的mRNA表达,Western印迹法测定各组大鼠心脏AT1受体的蛋白表达。结果与假手术组相比,心衰对照组左心室射血分数显著降低(P<0.05),而血浆PRA及AngⅡ水平升高(P<0.05),同时心脏AT1受体mRNA及蛋白的表达水平也显著上调(P<0.05)。给予姜黄素处理4周后,可明显改善心衰大鼠上述指标的异常(P<0.05)。而姜黄素对假手术组无相应作用。结论姜黄素通过降低心衰大鼠心脏AT1受体的表达,改善慢性心衰大鼠心功能。 Objective To explore the role of curcumin in angiotensin Ⅱ type- 1 receptor expression in rat hearts with heart failure. Methods Rat models of chronic heart failure (CHF) were established by left anterior descending coronary artery liga- ture. 32 Wistar rats were randomly divided into four groups: sham operation group, sham operation and curcumin treatment group, CHF model group and CHF model with curcumin treatment group. Cardiac function was examined by echocardiography after a 4 - week course of therapy. Plasma rennin activity (PRA) and angiotension II (Ang II) levels were tested by radioimmu- nity. The mRNA levels of AT1 receptor were measured by qRT - PCR. In addition, the level of cardiac AT1 receptor expression was detected by Western blotting. Results Compared with sham operation group, left ventricular ejection fraction (LVEF) of the CHF model group was significantly decreased (P〈0.05), while the plasma levels of PRA and Ang II were significantly increased (P 〈 0.05) and the expression of AT1 receptor mRNA and protein was up - regulated significantly (P 〈 0.05). After a 4 - week course of therapy, all of the above - mentioned abnormalities were reversed to some degree by curcumin treatment in the rats with heart failure (P 〈 0.05), but not in the rats of the sham operation group. Conclusions Curcumin reduces the expression of AT1 receptor in heart and improves heart function in rats with heart failure.
