摘要
目的 :探讨白细胞介素_10 (IL_10 )在牙周炎症反应中的抑制作用。方法 :用细胞培养法、酶联免疫吸附分析法(ELISA)检测细胞培养液上清中的IL_1β浓度 ;用MTT法检测人牙龈成纤维细胞 (HGF)对细菌脂多糖 (LPS)、IL_10的增殖反应。结果 :LPS以剂量依赖方式诱导HGF产生IL_1β,6h达最大分泌量 ;IL_1β的分泌可被IL_10抑制 ,10ng/mlIL_10作用 2 4h抑制效应达最大 ;LPS对HGF具有促增殖作用 ,而IL_10对HGF的增殖无影响。结论 :IL_10是一种抑制牙周炎症反应的细胞因子。
Objective:The purpose of this study was to investigate the inhibitory effect of interleukin_10 (IL_10) on the periodontal inflammatory reaction.Methods: Cell culture and ELISA were used to evaluate the concentration of interleukin_1β (IL_1β) in the supernatant, and MTT was used to examine the proliferation response of human gingival fibroblasts to lipopolysaccharides and IL_10.Results: IL_1β secreted by human gingival fibroblasts was induced by lipopolysaccharides in a dose_dependent manner and reached the highest level in 6 h. IL_1β was inhibited by IL_10, and the optimal inhibitory concentration of latter was 10 ng/ml. The inhibitory effect of IL_10 in the optimal concentration to IL_1β reached the maximum in 24 h. Further the proliferation of human gingival fibroblasts was also induced by lipopolysaccharides, but not affected by IL_10.Conclusion: IL_10 is a cytokine that can inhibit periodontal inflammatory reaction.\;
