银杏酮酯对抑郁大鼠行为及炎症细胞因子的影响 被引量:16

The effect of Ginkgo biloba extract 50 on behavior and inflammatory cytokines levels in depressed rats

摘要 目的:研究银杏酮酯(GBE50)对抑郁大鼠行为、血清和海马内炎症细胞因子含量的影响以探讨GBE50防治抑郁症的作用及机制。方法:将雄性SD大鼠随机分为6组,即空白对照组、模型组、氟西汀组(10mg/kg)以及GBE50(35、70和140mg/kg)组。采用孤养结合慢性不可预见性应激抑郁模型,每日给予一种应激刺激,共21d。在应激的同时每日灌胃给药,每周监测体重,造模结束后进行糖水偏爱度实验检测,之后腹主动脉取血、分离海马组织,ELISA法检测各实验组血清和海马炎症细胞因子的含量。结果:模型组体重明显低于空白对照组,各用药组体重与模型组之间没有明显区别,但GBE50各剂量组体重较模型组有增长的趋势。模型组糖水偏爱度明显高于空白对照组;与模型组相比,氟西汀组和GBE50(70和140mg/kg)组明显降低。模型组血清和海马内TNF-α、IL-1β、IL-6水平均明显高于空白对照组,血清IL-10水平明显低于空白对照组,而海马内IL-10水平明显高于空白对照组。与模型组相比,GBE50各剂量组血清TNF-α、IL-1β、IL-6水平以及氟西汀组血清TNF-α、IL-6水平均有不同程度降低;海马内GBE50(70和140mg/kg)组以及氟西汀组这些促炎症细胞因子以及IL-10的水平均明显下降。结论:抑郁大鼠体重明显减轻,表现出对糖水的异常偏爱,其血清和海马内促炎症细胞因子TNF-α、IL-1β、IL-6的含量升高,同时血清抗炎症细胞因子IL-10的含量降低,而海马内IL-10水平升高;GBE50可以明显改善模型大鼠抑郁样行为,逆转抑郁大鼠这些血清和海马炎症细胞因子的含量,这可能是GBE50抗抑郁作用的重要机制之一。 Objective:To investigate the effect of Ginkgo biloba extract 50 (GBES0) on behavior and inflammatory cytokines in depressed rats, some behavior tests and serum and hippocampal inflammatory cytokines levels were detected. Methods :The models were established by chro- nic unpredictable stress. Sixty male SD rats were randomly divided into six groups: Control group, model group, fluoxetine group (10mg/kg) and GBES0 groups (35, 70 and 140mg/kg). Body weight of rats was measured every week, after stress stimulate and oral administration for 21 days, sucrose preference test was measured, and serum and hippocampal levels of inflammatory cytokines such as TNF-ct, IL-113, IL-6 and IL-IO were detected by ELISA. Results: Model rats had obvious loss of body weight( P 〈 0.01 ), showed increased in sucrose preference compared to control rats( P 〈 0.01 ), and serum and hippocampal TNF-ct, IL-1 [3, IL-6 levels were obviously increased (P 〈 0.01 ). And IL- 10 level was significandy decreased in serum but increased in hippocampus in model group compared to control group( P 〈 0. 01 ). fluoxetine group and GBES0 three dose groups didnt show a significant difference in body weight with model group( P 〉 0.05 ), but all GBES0 groups showed the increasing trend. In sucrose preference test, Rats in fluoxetine group and GBES0 groups (70 and 140mg/kg) showed lower su- crose preference compared to model rats ( P 〈 0.05). In addition, serum TNF-α, IL-1 [3, IL-6 levels in GBESO three dose groups and TNF- α, IL-6 levels in fluoxetine group were significantly lower than that of model group, and serum IL-10 level in these GBES0 groups and fluoxe- tine group were obviously higher compared to. model group ( P 〈 0.05 - 0. 01 ). In hippocampus, TNF-ct, IL-1 [3, IL-6 and IL-10 levels in GBES0 groups (70 and 140mg/kg) and fluoxetine group were significantly decreased compared to model group( P 〈 0. 05 -0. 01 ). Conclu- sions:Depressed rats showed lower body weight and abnormal sucrose preference. Their serum and hippocampal pro-inflammatory cytokines TNF-α, IL-1 β, IL-6 levels were obviously increased, and anti-inflammatory cytokine IL-10 was obviously decreased in serum but increased in hippocampus, while GBESO can meliorate depression-like behavior and reverse these inflammatory cytokines levels in serum and hippocam- pus, these indicate that GBESO has anti-inflammatory effect and some degree of periphery and central immunoregulatory effect on depressed rats, which might be an imoortant antideoression mechanism.
