摘要
目的探讨柴芩承气汤对急性坏死性胰腺炎(ANP)犬肺巨噬细胞白细胞介素1β(IL-1β)表达的影响。方法将15只比格犬随机分成对照组、模型组及柴芩承气汤组各5只,模型组及柴芩承气汤组采用50 mg/kg牛磺胆酸钠逆行胆管注射制作ANP模型。造模成功后,对照组及模型组经胃管注入生理盐水10 ml/kg,柴芩承气汤组经胃管注入柴芩承气汤10 ml/kg,各组均2 h注入1次,共3次。第6 h取各组犬血清检测IL-1β水平,并取肺组织分离巨噬细胞检测IL-1βmRNA及蛋白表达。结果模型组、柴芩承气汤组肺组织巨噬细胞IL-1βmRNA表达水平均较对照组明显下调(P<0.05);模型组肺组织巨噬细胞IL-1β蛋白表达及血清IL-1β水平显著高于对照组,柴芩承气汤组IL-1β蛋白表达及血清IL-1β水平显著低于模型组(P<0.05)。结论柴芩承气汤可下调ANP犬肺组织巨噬细胞IL-1β蛋白表达,降低血清IL-1β水平。
Objective To observe the effect of Chaiqin Chengqi Tang on expression of interleukin-1β( IL-1β)in pulmonary macrophage of dogs with acute necrotic pancreatitis( ANP). Methods Fifteen beagle dogs were randomly assigned into control group,model group and Chaiqin Chengqi Tang group,with five dogs in each group.50 mg / kg of sodium taurocholate was retrograde injected through bile duct to establish ANP model. Dogs in control and model groups were administered with 10 ml / kg of saline via gastric tubes,while those in Chaiqin Chengqi Tang group were administered with Chaiqin Chengqi Tang in the same dosage and delivery route. Every group was administered one time per two hours,for three times. In the sixth hour,canine serum was collected for detecting interleukin-1β( IL-1β)and pulmonary tissue was obtained to isolate macrophages for detecting IL-1β mRNA and protein expression. Results IL-1β mRNA expression in pulmonary macrophages was significantly decreased in model and Chaiqin Chengqi Tang groups compared with that in control group( P〈0. 05). IL-1β protein and serum IL-1βexpressions in pulmonary macrophages were significantly higher in model group than those in control and Chaiqin Chengqi Tang groups( P〈0. 05). Conclusion Chaiqin Chengqi Tang can decrease the IL-1β protein expression of canine pulmonary macrophages,leading to the reduction of serum IL-1β.
