摘要
目的了解山东地区鼻咽癌(NPC)及健康人群中EB病毒(EBV)编码BARF0基因变异特征,探讨其变异意义。方法采用PCR和DNA测序检测NPC组织以及健康成年人咽漱液标本中EBV的BARF0基因序列,与EBV标准株进行比较,根据特征性突变对变异进行分类。结果 73例NPC病人和70例健康人群BARF0成功测序,共检出4处共有突变。其中36例(49.3%)NPC和29例(41.4%)健康人群同时出现4处共有突变(T160473G、T160545C、A160701C和C160707G),被分类为4M变异型;35例(48.0%)NPC和38例(54.3%)健康人群同时出现除T160473G外的3处共有突变,被分类为3M变异型,其余2例(2.7%)NPC和3例(4.3%)健康人群分类为Others。χ2检验精确概率法分析表明,BARF0变异型在2种人群中的分布差异无统计学意义(P=0.618)。结论山东地区NPC和健康人群中EBV毒株BARF0序列变异一致,BARF0基因变异的地域性分布及与EBV相关肿瘤的关系值得进一步研究。
Objective To investigate the characteristics of polymorphisms of BARF0 gene of Epstein-Barr virus(EBV)in nasopharyngeal carcinoma(NPC)and healthy crowd of Shandong province,and explore the significance of BARF0 gene variations. Methods Employing PCR and DNA sequencing,the BARF0 sequences of isolates from NPC biopsies and throat washing samples of healthy persons were detected.The sequences were compared with the EBV type strain,and the variations were classified according to distinctive mutation. Results A total of 143 isolates from 73 NPC and 70 healthy donors were successfully sequenced and four common mutations were detected.Thirty-six(49.3%)NPC and 29(41.4%)healthy donor isolates with four common mutations(T160473G,T160545 C,A160701Cand C160707G)were arranged into one group and named variant 4M,and35(48.0%)NPC and 38(54.3%)healthy donor isolates with three common mutations(T160545C,A160701 Cand C160707G)were characterized into one group and named variant 3M.The remaining two(2.7%)NPC and three(4.3%)healthy donor isolates were classified as"Others"group.Chi-square test showed that the distribution of the BARF0 variants among two population groups was not significantly different(P=0.618). Conclusion The isolates in NPC and healthy donors in Shandong province demonstrates consistent variation pattern.The geographical distribution of the BARF0 variants and the relationship between BARF0 variation and EBV-associated tumors are worth further study.
