摘要
目的:通过Box-Behnken效应面法优化川芎的提取工艺。方法:HPLC测定川芎饮片和药液中川芎嗪、阿魏酸、藁本内酯的含量,以川芎嗪、阿魏酸、藁本内酯转移率的多指标综合加权评分(以层次分析法确定其权重系数为0.6∶0.3∶0.1)为指标,通过Box-Behnken试验考察溶剂倍数、提取时间、乙醇浓度等因素的影响,对结果进行二项式方程拟合,利用效应面法优化提取工艺并进行预测分析。结果:川芎的最佳提取工艺为饮片浸泡1 h,以4倍量50%乙醇回流提取2次,每次2 h,预测值与验证值偏差较小。结论:该工艺稳定可行,实际值与预测值吻合度高,预测性良好,优选的工艺各成分提取率高,为川芎及其制剂的研究提供了实验依据。
Objective : To optimize extraction process of Ligusticum chuanxiong Hort by analytic hierarchy process and Box - Behnken design - response surface method. Method : The contents of ligustrazine, ferulic acid and ligustilide were de- termined by HPLC. Independent variable included concentraction of ethanol, liquid - solid ratio and extraction time and dependent variable included the muhi - index comprehensive weighted scoring method of extraction rate of ligustrazine, ferulic acid and ligustilide ( With analytic hierarchy process to determine the weight coefficient is 0.6 : 0.3 : 0.1 ). No - linear mathematic models were to estimate the relationship between independent and dependent variables and respone sur- face methodology was used to optimize extraction process and compare the observed and predicted values as prediction.; Results:Optimum estraction technology was listed as follows: Yinpian soak 1 h,4 times more than the volumn of 50% eth- anol solution,by heating reflux extraction for twice, each time 2 h. Deviation between the observed and predicted values was small. Conclusion:The optimized extraction technology was stable and feasible with high predictability, and it provides an experimental basis for compound preparation technology research of Ligusticum chuanxiong Hort.
