摘要
目的探讨槲皮素对不同时间和不同剂量染矽尘大鼠血清中硅(Si)、铜(Cu)、锌(Zn)元素水平的影响。方法采用正交设计,将108只无特定病原体级健康成年雄性SD大鼠随机分成对照组,矽肺模型组,以及按正交设计表L16(43)安排不同染毒剂量(分别为12.5、25.0和50.0 g/L)、不同染毒时间(分别为7、14、21和28 d)、槲皮素干预(分别为12.5、25.0和50.0 mg/kg)各水平组成的16个实验条件组。实验结束后,各组大鼠经心脏采血,采用电感耦合等离子体-质谱法测定血清中Si、Cu和Zn元素水平,并进行肺组织病理学检查。结果予槲皮素干预的大鼠肺泡炎及肺间质纤维化均较矽肺模型组有不同程度的减轻。矽肺模型组大鼠血清中Si、Cu元素水平和Cu/Zn比值均高于对照组(P<0.05)。不同剂量单纯染尘的正交8、11和14组大鼠血清Si元素水平均高于对照组(P<0.05)。不同剂量槲皮素干预的正交5、6、7、9、10和13组大鼠血清Si元素水平均低于矽肺模型组(P<0.05)。予槲皮素干预的正交5、13组大鼠血清Cu元素水平和Cu/Zn比值均低于矽肺模型组(P<0.05)。各组大鼠血清Zn元素水平分别两两比较,差异均无统计学意义(P>0.05)。结论槲皮素可以延缓实验性矽肺早期病变的形成和发展,Cu和Cu/Zn比值的变化可初步作为矽肺疗效的评价指标。
Objective To investigate the effect of quercetin on levels of silicon( Si),copper( Cu),and zinc( Zn) in serum of rats exposed to different doses and times of silica. Methods This study was based on the table of orthogonal design L16( 43) to analyze the effect alone. One hundred and eight specific pathogen free healthy male adult SD rats were selected and randomly divided into blank control group,silicosis model group,and 16 orthogonal groups were designed according to different dust exposure dose( 12. 5,25. 0,and 50. 0 g / L),different exposure time( 7,14,21,and 28 days) and quercetin intervention( 12. 5,25. 0,and 50. 0 mg / kg). The blood was collected from the heart and at the same time the pathological observation of lung tissue was conducted. Levels of Si,Cu,and Zn in rat serum were directly determined by inductively coupled plasma mass spectrometry. Results The pulmonary alveolitis and interstitial fibrosis in rats treated with quercetin were decreased in different degrees than the silicosis model group. The serum Si and Cu levels,and Cu / Zn ratio of silicosis model group were significantly higher than those of the blank control group respectively( P〈0. 05). The serum Si levels of the orthogonal 8,11,and 14 groups with different dose dust treatment were significantly higher than those of the blank control group respectively( P〈0. 05). Compared with silicosis model group,the serum Si levels of the orthogonal 5,6,7,9,10 and 13 groups were intervened by different dose quercetin were significantly lower respectively( P〈0. 05),while the serum Cu levels and Cu / Zn ratios of the orthogonal 5 and 13 groups were significantly lower respectively( P〈0. 05). No statistically significant difference was found in multiple comparison among the serum Zn levels of the totally 18 groups( P〉0. 05). Conclusion Quercetin can delay the formation and development of experimental silicosis in early pathological changes,and Cu and Cu / Zn ratio changes can be preliminarily used as the evaluation indexes for silicosis curative effect.
