玉米赤霉原生质体融合的研究 被引量:1

摘要 应用0.8%纤维素酶(来自Trichoderma.Pseudokoningii)及1.0%蜗牛酶(来自Achatina fulica Ferussac)的混合酶液制备玉米赤霉(Gibberella zeae)原生质体,以30%聚乙二醇(分子量4000)0.01mol/L CaCl_2溶液(pH7.0)作融合剂,进行了原生质体的营养互补融合及双亲紫外灭活融合。在四个菌株组成的两组配对体系中(701×801,7^(AT)×8^(BT))获得了稳定的二倍体及一株四倍体。融合体HL-5及Gl-1较其直接亲本具有良好的生理及遗传性状,二者的赤霉烯酮合成能力均接近高产亲本。 The protoplasts of F. graminearum were obtained by using a combined enzyme system containing 0.8% cellulase and 1.0% extract from Achatina fulica Ferussac digestied juice. The formation, regeneration and the fusion of protoplasts were studied. Protoplasts fusion of F. graminearum was performed by treating suspensions of protoplasts derived from two nutritionally complementing strains (701, 801) and two prototrophic strains(7AT, 8BT) with the fusogen containing 30% polyethylene glycol (M. W. 4,000), 0.01mol/L CaCl2, pH7.0 prior to fusion, the protoplasts of two prototrophic strains were irradiated by uv in order that the suvival may be about zero. The results of investigation for fusants indicated that HL-5 and GL-1 had good physiological and genetic characteristics. The Zearalenone productivity of the fusants was close to that of the high-producing parents.
