红细胞浓度对固相法血型鉴定的影响 被引量:2

Influence of erythrocyte concentration on solid phase method of blood group identification

摘要 目的:探讨红细胞抗原浓度对固相法ABO血型正定型及Rh(D)血型鉴定的影响。方法:随机选取健康成人O型Rh(D)阴性和AB型Rh(D)阳性个体各5例,采集静脉血3~5ml置于EDTA抗凝管中,将O型和AB型血液配制成不同高低浓度的红细胞悬液,并用固相法进行ABO血型正定型及Rh(D)血型鉴定,同时以10%浓度的红细胞浓度作为对照参考,观察假阳性和假阴性出现的情况及假性结果出现时的红细胞抗原浓度。结果:红细胞抗原浓度超过25%易出现假阳性;红细胞抗原浓度低于2%易出现假阴性;低浓度红细胞抗原对抗B、抗D的影响更明显;高浓度红细胞抗原对抗A的影响更明显。结论:红细胞浓度对对固相法ABO血型正定型及Rh(D)血型鉴定有影响,且高、低浓度对不同单克隆抗体结合抗原的能力影响不尽相同。 Objective:To investigate the influence of the concentration of red blood cell antigens on the solid phase method of ABO positive stereotypes and Rh(D)blood group identification.Method:Each five healthy adult with O Rh(D)negative and AB Rh(D)positive was randomly selected and 3~5ml blood was collected in EDTA anticoagulated blood collection tubes.The red blood cell suspensions with different high and low concentrations were prepared with O type and AB type blood,and detected using solid phase method of ABO positive stereotypes and Rh(D)blood group identification.While 10%concentration of red blood cell concentration as a control reference,false positive,false negative and the concentration of red blood cell antigens with false positive and false negative were observed.Result:More than 25% of the concentration of red blood cell antigens occurred false positives more easily and less than 2% more easily false negatives.The low concentrations of red blood cell antigens had more pronounced effect on anti-B and anti-D,and high concentrations had more obvious effect on anti-A.Conclusion:Erythrocyte concentrations may have effect on solid-phase method of ABO positive stereotypes and Rh(D)blood group identification,and different high and low concentrations may have different impact on the ability of different monoclonal antibodies bind antigen.
