摘要
[目的]建立肌卫星细胞和肌内前体脂肪细胞的共培养体系。[方法]用10μg/ml INS、1μmol/L DEX和115 ng/ml IBMX对北京油鸡肌肉SV细胞进行诱导后,通过油红染色法、细胞免疫学鉴定及关键基因的表达对诱导的SV细胞进行分析。[结果]油红O脂肪染色法鉴定结果表明,诱导后的细胞红色脂质密度较高,说明SV细胞经过诱导后含有更多的前体脂肪细胞。通过实时荧光PCR定量检测细胞发育的标志性基因,发现诱导与未诱导的肌肉SV细胞均会表达前体脂肪细胞标志性基因。[结论]肌肉SV细胞经过诱导后前体脂肪细胞的含量得到提高,有利于建立前体脂肪细胞和肌卫星细胞的体外培养体系。
[Objective] This research aimed to establish the co-cultures system of muscle satellite cells and intramuscular preadipocytes.[Method]The SV cells in the muscles of Beijing You Chicken were induced with 10 μg/ml insulin(INS),1 μmol/L dexamethasone(DEX)and 115 ng/ml 3-isobutyl-1-methyl xanthine(IBMX).And the induced SV cells were identified by oil red O staining,cellular immunological identification and the expression of key genes involved in adipogenesis and myogenesis.[Result] The results of oil red O staining showed that the induced SV cells contained higher-density red stained lipid,which indicated that the induced SV cells contained more preadipocytes.The key genes about the cell development were detected by real-time fluorescent quantitive PCR.The detection results showed that the key genes of preadipocytes could be expressed by induced and non-induced SV cells in the muscles.[Conclusion] The induced muscle SV cells contained more preadipocytes,which was be propitious to establish the co-culture system in vitro of muscle satellite cells and preadipocytes.
