摘要
目的探讨Heregulin、Erb B-2及MMP-7在大肠癌中蛋白和基因的表达及其相关性,并关注三者在大肠癌发生发展中的作用。方法以金华市中心医院收集的47例大肠癌根治术后新鲜标本为研究对象,用Western blot法检测Heregulin、Erb B-2在肠癌组织、癌旁组织及远癌组织的蛋白表达,用RT-PCR法检测Erb B-2在3组的mRNA表达;比较分析三者与大肠癌临床病理特征的关系。结果 Heregulin和Erb B-2在肠癌组织的蛋白表达较癌旁组织显著增多(P<0.01),而癌旁组织比远癌组织的表达也有所增加(P<0.05)。MMP-7在肠癌组织的表达较癌旁组织增多,差异有统计学意义(P<0.01),远癌组织未见表达;肠癌组织中Heregulin、Erb B-2及MMP-7的蛋白表达两两之间存在正相关关系。Erb B-2在肠癌组织的mRNA表达显著高于远癌组织(P<0.05)。结论Heregulin、Erb B-2及MMP-7在大肠癌中过度表达;Heregulin磷酸化激活Erb B-2后可能通过复杂的下游信号通路介导MMP-7表达上调,三者共同在大肠癌的浸润侵袭中发挥重要作用;Heregulin及Erb B-2有望成为判断大肠癌恶性进展的重要指标。
Objective To investigate the expression and correlation of Heregulin, ErbB-2 and matrix metalloproteinase-7 （ MMP-7 ） in carcinoma of large intestine, and evaluate their effects on the generation and progress of carcinoma of large intestine. Methods The fresh specimen of 47 cases carcinoma of large intestine after radical resection were collected. The proteins expression of Heregulin, ErbB-2 and MMP-7 in carcinoma tissue, adjacent non-tumor tissue and normal tissue surrounding cancer were determined by Western Blot. The levels of ErbB-2 mRNA in above three tissues were detected by RT-PCR. The relationship between Heregulin, ErbB-2 and MMP-7 and the clinicopathologic features of carcinoma of large intestine was discussed. Results The protein expression of Heregulin and ErbB-2 in cancer tissues increased when com- pared with the adjacent non-tumor tissue（P 〈 0.01 ） ,while their level in the adjacent non-tumor tissue was higher than that in the normal tissue surrounding cancer（ P 〈 0.05 ）. The expression of MMP-7 in cancer tissues was more active than the adjacent non-tumor tissue （P 〈 0.01 ）, but not in the normal tissue surrounding cancer. There was a positive correlation between any two of Heregulin, ErbB-2 and MMP-7 protein in carcinoma tissues. The level of ErbB-2 mRNA in cancer tis- sues was significantly higher than in normal tissue surrounding cancer. Conclusion Heregulin, ErbB-2 and MMP-7 are o- ver-expressed in carcinoma of large intestine. It is quite possible that Heregulin phosphorylation will activate ErbB-2, and then conducts the up-regulation of MMP-7 expression via complicated signal path. They play important roles in infiltration and attack of carcinoma of large intestine. The Heregulin and ErbB-2 is expected to become an important index to judge the malignant progress of carcinoma of large intestine.
