补肾法、疏肝法对超促排卵大鼠子宫内膜组织形态及血管生成的影响 被引量:19

Effect of Soothing Liver and Tonifying Kidney Method on Tissue Morphology and Angiogenesis of Endometrium in Superovulation Rats

摘要 目的观察补肾法、疏肝法对超促排卵大鼠妊娠结局的影响及可能作用机制。方法将80只动情周期正常的雌鼠随机分为正常组、模型组、补肾组、疏肝组各20只。除正常组外,各组大鼠建立控制性超促排卵模型。造模第1天始补肾组给予补肾调经方混悬液(浓度2.78 g/ml),疏肝组给予逍遥丸混悬液(浓度0.162g/ml),均1 ml/(200 g·d)灌胃,模型组及正常组给予同等剂量蒸馏水灌胃,连续11天。比较各组大鼠血清雌二醇(E2)、孕酮(P)含量及子宫内膜雌激素受体(ER)、孕激素受体(PR)水平,HE染色测量子宫内膜厚度并观察子宫内膜形态,观察大鼠内膜胞饮突表达,检测子宫内膜血管内皮生长因子(VEGF)蛋白及其mRNA表达,统计各组大鼠妊娠数、胚胎着床数。结果与正常组比较,模型组大鼠妊娠数、胚胎着床数减少,血清P含量及子宫内膜ER、PR蛋白表达,VEGF蛋白及其mRNA表达,子宫内膜厚度及螺旋动脉数量降低(P<0.05或P<0.01);与模型组比较,补肾组、疏肝组大鼠妊娠数及胚胎着床数增加,血清P含量及子宫内膜ER、PR蛋白表达,VEGF蛋白及其mRNA表达,子宫内膜厚度及螺旋动脉数量升高(P<0.05),而补肾组和疏肝组大鼠组间比较差异无统计学意义(P>0.05)。结论补肾法、疏肝法均能够增加超促排卵大鼠妊娠率及胚胎着床数,其作用机制可能与调节性激素水平及雌、孕激素受体,改善大鼠子宫内膜组织形态及血管生成有关。 Objective To observe the effect and possible mechanism of tonifying kidney and soothing liver on pregnancy outcome of superovulation rats. Methods Divide 80 female rats of normal estrous cycle into normal group ( n = 20) , model group ( n = 20) , tonifying kidney group ( n = 20 ) and soothing liver group ( n = 20 ) randomly. Except the normal group, the other 3 groups were built for controlled superovulation models. From the first day of modelling, tonifying kidney group was given suspension of Bushen Tiaojing Fang (补肾调经方, concentration of 2.78g/ml) , soothing liver group was given suspension of Xiaoyao Wan (逍遥丸, concentration of 0. 162 g/ml), both 1 ml/ (200 g· d ) by gavage, and the model and normal group were given distilled water of same dose by garage, for 11 days. Compare levels of estradiol ( E2 ), progesterone ( P), endometrium estrogen receptor (ER) and progesterone receptor (PR). Measure endometrial thickness by HE staining. Observe endometrial morphology and the expression of endometrial pinopodes. Detect endometrial vascular endothelial growth factor (VEGF) protein and the expression of mRNA. Count the number of encyesis and embryos implantation of each group. Results Compared with normal group, the number of pregnancy and embryos implantation, level of P in serum, the expression of endometrial ER, PR, VEGF protein and mRNA, endometrial thickness and spiral artery all decreased (P 〈 0.05 or P 〈0. 01 ) in the model group. Compared with model group, the number of pregnancy and embryos implantation, level of P in serum, the expression of endometrial ER, PR, VEGF protein and mRNA, endometrial thickness and spiral artery increased in the tonifying kidney group and soothing liver groups ( P 〈 0. 05 ). The difference was not significant between tonifying kidney group and soothing liver group (P 〉 0. 05 ). Conclusion Tonifying kidney and soothing liver could increase pregnancy rate and the number of embryos implantation of superovulation rats. The underlying mechanism may relate to regulating the sex hormone level, ER, PR and improving tissue morphology and angiogenesis in endometrium.
