偏头痛家兔模型的行为学评价方法

摘要 目的:探讨偏头痛家兔动物模型的复制方法,从行为学方面进行评价。方法:参照大鼠的偏头痛造模方法,按照5 mg/kg(低剂量)、7.5 mg/kg(中剂量)、10 mg/kg(高剂量)的标准进行颈部皮下注射硝酸甘油注射液,观察甩头、挠头、扒笼的行为学变化,同时检测血浆一氧化氮(NO)进行客观评价。结果:(1)低剂量组偏头痛家兔模型甩头次数和扒笼次数在31~120 min时段内、挠头次数在61~150 min时段内,较正常组均有明显差异(P〈0.01);(2)中剂量组偏头痛家兔模型甩头次数、挠头次数和扒笼次数在31~150 min时段内,较正常组均有明显差异(P〈0.05,P〈0.01);(3)低剂量组和中剂量组挠头次数在31~120 min时段内,比较均有明显差异(P〈0.05,P〈0.01);(4)三组不同剂量组的NO含量均较正常组高,与正常组均有明显差异(P〈0.05),三组间比较无明显差异(P〉0.05)。结论:5 mg/kg以上剂量均可诱发偏头痛发作,以7.5 mg/kg剂量较为适宜,注射后30 min起效,维持2 h,挠头是偏头痛家兔模型疼痛的突出表现,血浆NO升高是偏头痛家兔模型复制的评价指标。 Objective: To discuss the replication method of migraine rabbit models and to evaluate the behavior. Methods: With reference to the method of migraine rat models,nitroglycerin injection was given to the neck subcutaneous according to the standards of 5 mg / kg(low dose),7. 5 mg / kg(medium dose) and 10 mg / kg(high dose). We observed the changes of behavior;glancing,scratching the head and grilled cage. At the same time,we objectively evaluated nitric oxide(NO) in the plasma. Results:(1) The numbers of glancing and scratching the head of low dose group of migraine in the period of 31 - 120 min and the numbers of grilled cage in the period of 61 - 150 min had a very significant difference compared with those of the normal group(P〈0. 01).(2) The numbers of glancing and scratching the head of medium dose group of migraine in the period of 31 ~ 120 min and the numbers of grilled cage in the period of 61 - 150 min had a very significant difference compared with those of the normal group(P〈0. 05,P〈0. 05).(3) The numbers of scratching the head in the low dose group in the period of 31 - 120 min had a very significant difference compared with that in the medium dose(P〈0. 05,P〈0. 01).(4) The NO content of three different doses groups were higher than that of the normal group and there was no significant difference among the three groups(P〈0.05). Conclusion: It can induce migraine attacks to give nitroglycerin injection more than 5 mg / kg dose and the appropriate dose is7. 5 mg / kg. The effect starts after 30 min and maintains 2 hours. Scratching the head is the outstanding performance of the migraine rabbit models. The rise of NO in the plasma is the evaluation index of migraine rabbit models replication.
