摘要
为了给胰蛋白酶为主要成分的药物在奶牛体内的药代动力学提供合适的研究方法,以体外添加胰蛋白酶的奶牛血清和牛奶为研究对象,优化商品化的胰蛋白酶活性检测试剂盒,采用紫外分光光度法检测奶牛血清和牛奶中胰蛋白酶的活性。结果显示:0~0.4 nmol·m L-1范围内,胰蛋白酶的吸光度值与浓度呈现良好的线性关系,回归方程y=0.023x-0.003 8,r2=0.999 6;以信噪比S/N=3为标准,在牛奶中最低检测限浓度为25 ng·m L-1,平均回收率为84.3%,3 d中日内变异系数分别为13.9%、10.2%、11.8%,日间变异系数为11.0%;在血清中最低检测限浓度为15 ng·m L-1,平均回收率为85.6%,3 d中日内变异系数分别为13.7%、13.2%、8.1%,日间变异系数为13.8%。结果表明:该方法检测牛奶和血清中的酶活准确率高,在试剂盒可检测的浓度范围内受牛奶和血清中的蛋白成分影响很小,且回收率和精密度也都达到了兽用药物药代动力学试验指导原则的要求,具有良好的准确性和重现性。
The aim of this study is to provide an appropriate method for in vivo pharmacokinetics of trypsin in the drugs. Trypsin was added to the serum and milk and its activity was determined by the optimized detection kit. The results showed that there was a good linear relationship between the optical density and standard concentration of trypsin during the range of 0 ~ 0. 4 nmol·m L-1,with regression equation y=0. 023x-0. 003 8,r2= 0. 999 6. The lowest detectable limit of trypsin in the tested milk was 25 ng·m L-1. The average recovery in the milk was 84. 3%. The RSD of inner-day in the milk was 13. 9%,10. 2% and 11. 8%,respectively. The RSD of intra-day in the milk was11. 0%. The lowest detectable limit of trypsin in the serum was 15 ng·m L-1. The average recovery in the serum was 85. 6%. The RSD of inner-day in the serum was 13. 7%,13. 2% and 8. 1%,respectively. The RSD of intra-day in the tested serum was 13. 8%. The improved method was tested to have a high sensitivity,good accuracy and precision.
