摘要
目的:观察慢性束缚应激肝郁脾虚证模型大鼠中枢海马、杏仁核和外周胃、结肠组织P物质(substance P,SP)、血管活性肠肽(vasoactive intestinal peptide,VIP)mRNA表达的变化及逍遥散对其影响。方法:将30只雄性SD大鼠按随机数字表法分为正常对照组、模型组、逍遥散组,以慢性束缚应激方法制作肝郁脾虚证大鼠模型并用逍遥散进行干预。造模21 d结束后,采用实时荧光定量PCR方法检测海马、杏仁核、胃、结肠组织SP、VIP的表达。结果:与正常对照组比较,模型组大鼠海马、结肠组织SP表达显著升高(P<0.01),模型组大鼠VIP表达在海马显著升高(P<0.01)、杏仁核和胃组织表达显著降低(P<0.01或P<0.05);逍遥散能显著降低模型大鼠海马、结肠组织SP及海马VIP表达,升高杏仁核和胃组织VIP表达(P<0.01或P<0.05)。结论:脑肠肽SP、VIP在中枢和外周的分布异常可能是肝郁脾虚证脑肠互动功能发生障碍的原因之一,逍遥散对中枢和外周SP、VIP均有调节作用。
Objective: To investigate the effect of Xiaoyao powder decoction on substance P (SP) and vasoactive intestinal peptide(VIP) mRNA expression in the hippocampus, amygdala and stomach, colon of rats with stagnation of liver qi and spleen deficiency syndrome induced by chronic immobilization stress. Methods: 30 male SD rats were randomly divided into normal control group, model group, Xiaoyao powder group. Except for those in the normal control group, all the rats were subjected to chronic immobilization stress for 21 consecutive days and intervened with xiaoyao powder, the expression of SP and VIP in hippocampus, amygdala and stomach, colon were detected by real-time quantitative PCR method. Results Compared with normal control group : in the model group rats , expression of SP in hippocampus and colon was significantly increased (P 〈 0.01 ) , expression of VIP in hippocampus was significantly increased ( P 〈 0.01 ) , but decreased significantly in amygdala and stomach ( P 〈 0. 01 or P 〈 0. 05 ) ; Xiaoyao powder decoction could significantly decrease the model rats hippocampus, colon SP and hippocampus VIP mRNA expression, increase amygdala and stomach VIP mRNA expression(P 〈0. 01 or P 〈0. 05). Conclusion:Gbrelin SP and VIP distribution in the central and peripheral anomalies may be one of the reasons for brain-gut interactive function obstacles of stagnation of liver qi and spleen deficiency syndrome,Xiaoyao decoction have a regulatory role both on the central and peripheral SP and VIP.
