黄柏与抗菌药联合抗耐药大肠杆菌的作用研究 被引量:11

摘要 为研究黄柏水提取物与抗菌药联合体外抗含fos A3和β-内酰胺酶(ESBLs)型耐药基因大肠杆菌的效果,对临床分离的细菌进行鉴定,确定细菌所含耐药基因种类,采用二倍微量稀释法分别检测黄柏水提取物与抗菌药物的最小抑菌浓度(MIC),并以1/2 MIC的黄柏水提取物与抗菌药联合抑制耐药大肠杆菌。结果显示,分离到含fos A3和CTX-M耐药基因大肠杆菌;黄柏水提取物的MIC大于0.5 g/m L,以1/2 MIC的黄柏水提取物与14种抗菌药联合使用3代后,抗菌药抗菌活性显著增强,黄柏水提取物可显著降低抗菌药MIC,头孢曲松钠、阿莫西林、环丙沙星、左旋氧氟沙星、黏杆菌素、痢菌净、磺胺间甲氧嘧啶、加替沙星、头孢他啶、头孢噻呋钠、氟苯尼考、磷霉素的MIC均降到1.562 5μg/m L以下。黄柏水提取物联合抗菌药应用可能具有协同抑菌作用,可用于耐药细菌感染疾病的临床治疗。 The antibacterial activity of water extracts of Amur Corktree Bark combined with antibacterial agents were tested against the fos A3 and extended-spectrum β-lactamases( ESBLs) producing Escherichia coli. Bacterias isolated were identified and genes were confirmed by PCR amplification. The minimal inhibitory concentration( MIC) of water extracts of Amur Corktree Bark and antibacterial agents were tested through twice micro-dilution method. The bacterial was inhibited three generations by the 1 /2MIC of water extracts of Amur Corktree Bark combined with antibacterial agents. The result showed that the bacteria isolated was E. coli containing fos A3 and CTX-M gene. The MIC of water extracts of Amur Corktree Bark was above 0. 5 g / m L. The antibacterial activity of antibacterial agents were strengthened and the MIC of antibacterial agents were reduced significantly,MIC values of many antibacterial agents were below 1. 5625 after three generations of the 1 /2MIC water extracts of Amur Corktree Bark combined with 14 antibacterial agents separately,such as ceftriaxone sodium,amoxicillin,norfloxacin,ciprofloxacin,gatifloxacin,levofloxacin,colymycin,mequindox,sulfamonomethoxine,ceftazidime,ceftiofur sodium,fosfomycin. Diseases could be treated effectively by the water extracts of Amur Corktree Bark combined with antibacterialagents which caused by drug-resistant bacteria.
