摘要
目的探讨灵芝孢子粉抗抑郁作用机制。方法造模方法为慢性轻度不可预见性的应激模型,将SD大鼠随机分成5组,即空白对照组、模型组、盐酸氟西汀组(20mg/kg)、灵芝孢子粉高、低剂量组(50,100mg/kg),造模同时ig给药,每日1次,连续给药28d,均按10.0ml/kg给药,对照组模型组ig等量蒸馏水,观察大鼠行为学,采用酶联免疫法检测大鼠血清炎症因子(IL-6、TNF-α),采用LC-MS法测定大鼠脑内去甲肾上腺素(NE)、5-羟色胺(5-HT)含量,采用Westernblot检测大鼠海马区神经营养因子BDNF蛋白表达。结果灵芝孢子粉能显著改善抑郁大鼠行为学,能显著减少抑郁大鼠血清中的炎症因子(IL-6、TNF-α)的含量,能减少大鼠脑内NE的含量,增加5-HT的含量,并能增加抑郁大鼠脑内海马区BDNF的蛋白表达。结论灵芝孢子粉具有抗抑郁作用。
Objective To investigate the effects of anti - depressive mechanism of Ganoderma lucidum spore powder. Method- s using the chronic unpredictable mild stress to hulid depressive model, SD rats were randomly divided into 5 groups, namely control group, model group, fluoxetine group (20mg/kg) , Ganoderma lucidum spore powder high and low dose group (50mg/ kg, 100mg/kg) ,all drugs are administrated by Ig, one tome a day, continuous 28d, according to the 100mL/kg administration, control group, model group Ig distilled water, observe the behavior of rats, using enzyme - linked immunosorbent assay to detect inflammatory cytokines ( IL - 6, TNF - a) in serum, using LC - MS method to detect norepinephrine ( NE), 5 - hydroxytryptamine (5 -HT) content in brain, using the WESTERN BLOT to detect neurotrophic factor BDNF protein expression in hippoeampus of rats. Results Ganoderma lueidum spores can significantly improve the depressive behaviors of rats, reduced the inflammatory cytokines (IL -6, TNF - a) content, reduce the content of NE in brain, increased the content of 5 - HT, and increased BDNF in hippocampus. Conclusion Ganoderma lucidum spore powder has antidepressant effect.
