摘要
目的探讨检测脓毒症大鼠血清中miR-9的表达及其意义。方法将54只大鼠分为假手术组24只、脓毒症组24只和正常对照组6只。建立盲肠结扎穿孔脓毒症动物模型。检测各组大鼠血清中白细胞介素-6(IL-6)、肿瘤坏死因子α(TNF-α)和C反应蛋白(CRP)浓度水平,评估脓毒症炎症反应情况;实时荧光定量RT-PCR法检测大鼠血清中miR-9的表达情况;皮尔森相关分析评估血清miR-9的表达与IL-6、TNF-α和CRP之间的关系。结果脓毒症大鼠血清miR-9的表达水平较正常对照组和假手术组显著升高(P均<0.01),术后2 h血清miR-9表达即迅速上升达到一个较高水平。皮尔森相关分析结果也显示血清miR-9的表达与IL-6、TNF-α和CRP呈正相关(P均<0.01)。结论脓毒症大鼠血清miR-9表达显著增高,其水平反映机体炎症反应状态,可以作为脓毒症早期诊断及预后的指标。
Objective It is to investigates the expression of miR- 9 in serum of septic rats,as well as its significance.Methods 54 rats were divided into sham group( n = 24),sepsis group( n = 24) and normal control group( n = 6). Animal models of sepsis were established by cecal ligation and puncture. The serum levels of IL- 6,TNF- α and CRP were measured in every group to evaluate the severity of sepsis. The expression level of serum miR- 9 was determined by reverse transcription polymerase chain reaction( RT- PCR). Pearson's correlation analysis was used to evaluate if the expression of serum miR-9 had correlation with IL- 6、TNF- α and CRP. Results Expression level of miR- 9 in rats' serum in sepsis group was significantly higher than that in normal control group and sham group( P〈0. 01),which was very low under normal condition,was significantly increased at 2 h after CLP. Pearson's correlation analysis also showed that the level of serum miR- 9 had positive correlation with IL- 6、TNF- α and CRP( P〈0. 01). Conclusion The significantly up-regulated expression levels of serum miR- 9 in septic rats reflected the situation of inflammatory response,and may be used as a diagnostic and prognostic marker in sepsis.
